Objective:To investigate the effects of Stichopus japonicas acidic mucopolysaccharide(SJAMP) on PCNA expression and cell cycling in invitro cultured Henrietta Lacks strain of cancer cells(Hela cells) and compare these with methotrexate(MTX).Methods:The cell cycle was measured using flow cyt ometry(FCM),and the level of proliferating cell nuclear antigen(PCNA)with immunohistochemistry(IHC).The results were compared between Sjamp and MTX.Results:Both Sjamp and MTX led to significantly suppressed expression of PCNA as well as ahered cell cycling in Hela cells.Compared with no- intervention group,more cells in the intervention group were trapped at G1 phase(P<0.05)and fewer caught at S and G2 phases(p<0.05).Level of PCNA of Sjamp-treated and MTX-treated groups declined significantly compared with controls(P <0.05),but these changes did not differ statistically between the two groups.Conclusion:The findings suggested inhibited PCNA expession and retarded cell cycle in Hela cells treated with Sjamp was similar to those with MTX.
