Objective: To compare the effects of Matrine and Methotrexate(MTX) which used primarily in rheumatoid arthritis(RA) on proliferation, expression of PCNA and cell cycle in Hela cells by using cell culture in vitro.Methods: To compare the effects of different concentration of Matrine and MTX on proliferation of Hela cells by MTT, to measure the expression of PCNA in different concentration group by histochemistry and the cell cycle in each group by FACS.Results:â‘ Both Mat and MTX have an inhibitory effect on the proliferation of Hela cells invitro, which was dose and time dependent, the inhibitory rate increase with the increase of the concentration. Compared MAT, MTX group with the control group, there was statistical significance(P<0.05), but there was no significance between the MAT and MTX group(Pï¹¥0.05).â‘¡Compared with the control group, there was statistical significance in the expression of PCNA among different concentration of MAT and MTX. The mean absorbance decreases with the increase of MAT and MTX concentration. But there was no significance between MAT and MTX group. It showed that the effect of MAT and MTX on the expression of PCNA has no statistical difference within the selected range of concentration(Pï¹¥0.05).â‘¢The difference of cell cycle and mean of PI% between MAT and MTX group was not statistically significant in three levels of MAT and MTX. But there was statistical significance compared MAT and MTX group with control group(P<0.05). MAT and MTX was able to decrease the cells of S stage, increase cells of G0/G1, besides PI% lowered too compared with control group.Conlusion: There was no statistical significance between MAT and MTX in their effects on proliferation, expression of PCNA and cell cycle in Hela cells within the appropriate concentration range. The MAT and MTX has a similar mechanism so that there may be a therapeutic prospect of MAT in RA.
