Objective:To establish an animal model of Oxygen-Induced Retinopathy of prematurity and inject Avastin into vitreous cavity o Examining the expression of MMP-9 in retinal tissue to explore the Treatment of Avastin in Oxygen-Induced Retinopathy of prematurity in the mouse。Methods:To establish an animal model of Oxygen-Induced Retinopath of prematurity。One handred and twenty SD mice aged 7days were devided into four groups that include normal group,hyperxia group,treatment group and control group。The last two groups were further devided into three groups that include 1μl group,5μl group and 10μl group。The expression of MMP-9 protein was assessed by immunocytochemistry assay,and the MMP-9 mRNA was studied by reverse transcriptase PCR。Results:MMP-9 was detected by immunocytochemistry assay in normal retinal tissue, and its expression increased in hyperxia group(P<0.01)。The changes of MMP-9mRNA paralleled to the changes of its protein。The expression of MMP-9 in the three treatment groups were 134.34±4.42,137.77±6.43,142.97±6.31,and the MMP-9 mRNA were 0.8067±0.05,0.7183±0.06,0.5537±0.040 Compared to the hyperxia group,its expression decreased(P<0.01)。The expression of MMP-9 between hyperxia group and control group has no difference(P>0.05).Conclusion:The expression of MMP-9 were increased in Oxygen-Induced Retinopathy of prematurity。Avastin could down-regulate the expression of MMP-9。So Retinal neovascularization can be inhibitted by intravitreal injection of Avastin。...
