| Objective:To study the express of VEGF in the vitreous humor and retina,and the effect of avastin on the content of VEGF in the vitreous humor and retina. Methods:To select 160 seven days old SD mice,divided them into normal control group,high-oxygen-induced retinopathy group,treated group and balanced-salt-solution(BSS) group,the last two groups were divided into large,middle and little dosage groups.Except for the control group,the mouse of another groups were taken into the high-oxygen box five days with the oxygen concentration of(75±2)%,then all of the mice were taken in the air for five days.As soon as took them into air,the treated group were injected with 1μl,5μl,10ul avastin,BSS group were injected with 1μl,5μl,10μl BSS in the vitreous except the control group and high-oxygen-induced retinopathy group.32 eyes in all the 40 eyes of these mice in every group were punctured and taken vitreous humor.We measured the concentration of VEGF in their vitreous humor by means of enzyme-linked immunosorbent assay method.Another 8 eyes were put into formaldehyde with the concentration of 10%,then we scaled wax, cut sheet,detected the expression of VEGF in the retina of all groups with immunohistochemistry.Results:the content of VEGF in the retina and vitreous humor of high-oxygen-induced retinopathy group mice were more than that of the normal control group (P<0.01),As the content of avastin increased,the content of VEGF in retina and vitreous humor of treatment group reduced(P<0.05).Conclusion:the animal model of oxygen-induced retinopathy was successful.the content of VEGF in the retina and vitreous humor of oxygen-induced retinopathy mouse was more than that of control group.avastin could degrade the content of VEGF.with the concentration increasing,its effect would be great. |
