Objective: To investigate the effect of smoking and smoking cessation on glycometabolism, lipometabolism and oxidative stress in type 2 diabetes rats.Methods: 42 six-weeks-old Wistar rats were randomly divided into 2 groups: normal(n=21) and diabetes(n=21). Each group was assigned randomly into control 8 weeks subgroup(n=7), smoking 8 weeks subgroup(n=7) and smoking cessation 8 weeks subgroup. Smoking groups were given passive smoking twice daily for 8 weeks and smoking cessation groups giving up smoking for 4 weeks after 8 weeks smoking. Then the concentrations of malondialdehyde(MDA), and the activity of antioxidant enzymes including superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in serum and tissues (liver, kidney) were measured, respectively. Other diabetic related pathophysiologic parameters such as fasting insulin, blood glucose and blood lipid were observed as well, respectively. Results: DM group showed a significant increasing of blood glucose, total cholesterol, triglyceride compared with NC group, respectively. The concentrations of MDA in serum and tissues of liver and kidney of DM group were increased significantly compared with NC group, respectively, while the activity of SOD and GSH-Px decreased significantly. The concentrations of MDA in serum and tissues of liver and kidney of smoking group were increased significantly compared with control group, respectively, while the activity of SOD and GSH-Px decreased significantly. And smoking cessation may reverse the above changes.Conclusion: These data confirm that there was severe oxidative stress state existed in diabetic rats. Oxidative stress probably plays an important role in the development of diabetes and its chronic complications. Smoking may increase the oxidative stress in diabetic rats, thus affecting the occurrence and the development process of diabetes, but to quit smoking can reduce the body's oxidative stress and conducive to the improvement of diabetes.
