Objective:To investigate the effect of smoking and smoking cessation on the NF-κB p65 expression in rat skeletal muscle nuclei.To investigate whether smoking through the effects of NF-κB p65 expression affected the occurrence and development of diabetes,and whether smoking cessation can reverse the above changes.Methods:84 rats,Were randomly divided into control group,diabetes mellitus group.And each group were randomly divided into six groups: non-smoking 8-week group,non-smoking 12-week group,smoking 8-week group,smoking 12-week group,smoking cessation 8-week group,smoking cessation 12-week group.STZ(Sigma,USA) were administered intraperitoneally at a dose of 33 mg/kg after 4 weeks of high sugar high fat diet.After 72 hours,animals with FBG≥16.7mmol/l were served as diabetic models.The animals were respectively executed in 8 weeks,12 weeks,16 weeks, fasting blooding samples were collected for the assessment of FBG,FINS,TNF-α,IL-6,TC,TG,HDL,LDL,FFA levels.Western blot analysis NF-κB p65 in skeletal muscle nucle.Results:(1)In smoking 8-week diabetes group p65 expression is higher than in non-smoking 8-week diabetes group,In smoking 12-week diabetes group p65 expression is higher than in non-smoking 12-week diabetes group. (2)In smokingl2-week diabetes group p65 expression is higher than in smoking 8-week diabetes group.(3)In smoking cessation 8-week diabetes group p65 expression is lower than in smoking 8-week diabetes group,In smoking cessation 12-week diabetes group p65 expression is lower than in smoking 12-week diabetes group.(4)In smoking cessation 8-week diabetes group p65 expression is higher than in non-smoking 8-week diabetes group,In smoking cessation 12-week diabetes group p65 expression is higher than in non-smoking 12-week diabetes group.Conclusions:Smoking promote NF-κB p65 expression in diabetic rat skeletal muscle nuclei,with the time and smoking,NF-κB p65 expression is higher,smoking cessation can be partially reversed the above changes.
