[OBJECTIVE] To Set up a model of Rat type 2 diabetes and to explore the significance of fgl2 and 6-keto-PGF1α/TXB2 imbalance in the coronary microangiopathy of the model.[METHODS] The Sprague-Dawley male rats were randomly assigned to the model group(n=50) and the normal group(n=30) in this study.After two months'high fat diet,STZ was injected into its abdominal cavity in the model group,36 Rats were up to standard,of which 6 died of the disease.The normal group was given full diet.It was observed that the heart ,cornory microvascular,the expression of fgl2 and the level of 6-keto-PGF1αTXB2 in the blood by HE stain,MSB,immunostaining and radio-immunity.[RESULTS]①HE and MSB stain showing:there was homogen pink thrombus-like substance in part of vasculars under endocardium in the model 2m,3m,4m group,which was positive in MSB stain.Microvascular wall was thicken . Myocardial cell was shrinked.The fiber was vague and ranked disordered. Fibroplasia could seen between the cells.The normal group was negative.②Immunostaining showing:fgl2 was specificly expressed on the microvascular wall in the model 2m,3m,4m group. There was significant deviation between the subset (р<0.01) .The normal group was negative.③The level of 6-keto-PGF1αTXB2 in the blood : the level of 6-keto-PGF1αwas decreasing with the course .There was significant deviation between the model 2m,3m,4m group and the control(р<0.05,р<0.01),while TXB2 was increasing andр<0.05.④The correlation analysis between fgl2 and 6-keto-PGF1αTXB2 :there was significant negative correlation between fgl2 and 6-keto-PGF1α(r=-0.528,р<0.01),while significant positive correlation between fgl2 and TXB2(r=0.781,р<0.01).[CONCLUSIONS]①Microvascular hyaline thrombosis and microangiopathy could seen in the heart of Rat DM.②The expression of fgl2 and the imbalance of 6-keto-PGF1α/TXB2 could observed in the heart of Rat DM.And there was significant correlation between them.③The expression of fgl2 and the imbalance of 6-keto-PGF1α/TXB2 could be in coordination with each other in the development of the heat microangiopathy of DM.
