Objective: By observing the Fgl2 enzyme prothrombin in the peripheral blood mononuclear cells PBMCs) and serum inflammatory cytokines TNF-αexpression level in patients with acute myocardial infarction(AMI),and comparing with the ones expressed in patients with stabled angina pectoris (SAP) and the normal control group to investigate how the Fgl2 and TNF-αtakes part in the immune coagulation mechanism of acute myocardial infarction .Methods: Select patients from the Union Medical College Hospital in-patient departments Cardiovascular medicine as research objects , including acute myocardial infarction (AMI) 29cases, stable angina pectoris group (SAP) 28 cases, normal voluntary (CG) 18 cases. we got blood sample in vein from all these people ,who had shown full understanding and permission to our study in advance .The sample obtained from Acute myocardial infarction (AMI)groups at acute onset in 24h and from the stable angina group during the both admitted to hospital immediately . The control group sample obtained randomly. All patients and normal people never used anticoagulants or thrombolytic therapy. Respectively, we detect the Fgl2 protein expression levels in peripheral blood mononuclear cells (PBMC) by immunofluorescent technique and westen-blot test; and measure the cytokines concentration of TNF-αin the peripheral blood Serum by ELISA in each group of people.Results: 1.The immunofluorescent test showed that the average optical density of Fgl2 expression in Acute myocardial infarction(AMI)group was significantly higher than that of in stable angina group and in the control group .The difference was statistically significant (P <0.05). 2. ELISA results showed that the concentration of TNF-αin patients with acute myocardial infarction was significantly higher than stable angina and the control group(76.20±5.79 vs 38.54±1.98,35.92±2.03, P<0.05).There was no significant difference between the stable angina group and the control group ( 38.54±1.98 vs35.92±2.03 P>0.05).3.the results of westen-blot showed the concentration of Fgl2 expressed in the control group was few and almost negative . while , which expressed in SAP group was low and in AMI group was very high .Conclusions: 1.The Fgl2 expression level in peripheral blood mononuclear cells from Acute myocardial infarction patients was significantly increased, so we can deduce that it may be one of the coronary Micro thrombosis acute-phase protein and may have the predictive value.2.The increases of serum levels of inflammatory factors in Acute myocardial infarction patients suggest that inflammation initiate or promote the development of acute myocardial infarction. Objective:By observing the Fgl2 enzyme prothrombin in the peripheral blood mononuclear cells PBMCs) and serum inflammatory cytokines TNF-αexpression level in rats heart ischemia reperfusion. model ,to discuss the importance and the relationship between the FGL2 and TNF-αduring ischemia reperfusion.Methods:56 male SD rats were randomly divided into three groups,control group( n=8 ) ,sham-operated group (n=24 ) and operation-treated group ( n=24 ) .The operation-treated group was divided into three subgroups ,the ischemia reperfusion 1 day group,the 3days group and the 7days group.the rats were killed in due time ,the samples were obtained for test .Serum cytokines TNF-αwere tested with the ELISA kits.Fgl2 expression in the microvasculatures were detected by Westen -blot test.Results: 1.The results of ELISA test showed the concentration of TNF-αin operation-treated group was much higher than that in control group and sham-operated group (40.68±6.73,66.36±4.05,67.12±3.88 vs 30.78±6.24,27.36±6.20, P<0.05).There is no significant different between sham-operated group and control group ( 30.78±6.24 vs27.36±6.20 P>0.05 ) .2.The westen-blot test showed that the expression of microvasculature fgl2 in the control group and sham-operated group was very low almost negative ,while ,which in the operation-treated group was very high,and the concentration in the 3days group and 7days group both higher than the 1day group . Conclusions:1.The results showed that there was micro thrombosis in coronary micro-vessel in rats ischemia reperfusion models ,and the microvasculature fgl2 expression was increased ,which may be one of the the immune coagulation mechanisms to micro circulatory disturbances after ischemia reperfusion process ,and may also be one of the causes of formation to the micro thrombosis .2. After ischemia reperfusion ,the expression of Fgl2 and the concentration of TNF-αwere both increased ,and in the same trend ,so we can deduce that there must be some correlation with the fgl2 and TNF-αduring the formation of micro thrombosis.
