Objective: To observe the endotoxemia initiated inflammation and immune function in endotoxin tolerant rats.Methods: Ninty SD male rats were divided up into three groups randomly: normal control group (30 rats),LPS-treated group(30rats) and endotoxin tolerant group(30rats). Endotoxin tolerance was induced by intraperitoneal injection of 0.25 mg/kg Escherichia coli LPS (serotype 0127:B8) at the first day and intraperitoneal injection of 0.5mg/kg LPS on days 2-5. LPS-treated rats received an equivalent amount of saline. 72 hours after the last LPS injection, LPS-treated rats and endotoxin tolerant rats were intraperitoneal injected with 10mg/kg of LPS. The levels of sIgA in intestinal mucosal were measured by radioimmunoassay in LPS-treated and endotoxin tolerant rats after intraperitoneal injection of 10mg/kg lps 0,2,4,6,12 hours. Immunohistochemistry of ICAM-1 in the intestine was measured. RESULTS: Weight of ET rats didn't reduce after injected 0. 5mg/kg LPS. The levels of sIgA in intestinal mucosal of ET rats were higher than that of normal rats, especially at 6 hours (P<0.05). The levels of sIgA of LPS-treated rats began to reduce from 6 hours (p<0.05).The expression of ICAM-1of ET rats did not rise, and the expression of ICAM-1 was lower compared with the LPS-treated group.(p<0.05) But in LPS-treated group, it gradually rised. From histopathologic sildes, there was more obviously mucosa congestion,edema in the intestinal mucosa and with a lot of inflammatory cells in LPS-treates group than endotoxin tolerant group. Chiu pathologic grade was lower in endotoxin tolerant group compared with the LPS-treated group.Conclusions: Endotoxin tolerance can reduce intestinal mucosal injury and the expression of ICAM-1 caused by endotoxemia.
