Partâ… Repression of vascular endothelial growth factor by Thalidomide in human nasopharyngeal carcinoma CNE-2 cellsObjective: To evaluate the effect of Thalidomide on the growth and the expression of vascular endothelial growth factor (VEGF) in human nasopharyngeal carcinoma cell line CNE-2.Methods: Cell growth inhibition was assessed by MTT-microculture tetrazolium assay after incubated with Thalidomide at various concentrations and times; the VEGF and VEGF-C mRNA and protein expression in CNE-2 cells were detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry respectively.Results: Growth of CNE-2 cells was inhibited by Thalidomide in a dose-dependent and time-dependment manner, and the inhibitive rate was 45.64% after incubation with 60mg/L Thalidomide for 72 h. The VEGF and VEGF-C mRNA expression levels in CNE-2 cells were all reduced after treatment of Thalidomide, especially the VEGF mRNA. Their protein expression levels were consistent with their mRNA expression levels.Conclusions: Thalidomide can inhibit the growth of CNE-2 cells, which may be related to its effect of down-regulating VEGF expression. Partâ…¡Effect of Thalidomide on the radiosensitivity in vitro in human nasopharyngeal carcinoma cell line CNE-2Objective: To evaluate the effect of Thalidomide on the cell cycle and the radiosensitivity in human nasopharyngeal carcinoma cell line CNE-2.Methods: CNE-2 cells were exposed to Thalidomide or/ and irradiation, the cell survival curves and SERs ( sensitive enhancement ratio ) were obtained with the colony formation assay; cell-cycle kinetics was analyzed by flow cytometry(FCM).Results: Thalidomide of 20mg/L and 40mg/L both had radiosensitive effect on CNE-2 cells exposed for 24 h, and SERs of which were 1.29 and 1.57; When CNE-2 cells were treated with different concentrations of Thalidomide for 24 hours, it showed that the G0/G1 cells increased, the S and G2/M cells decreased, but no significant differences were found in cell-cycle.Conclusions: Thalidomide can increase the radiosensitivity.
