Objective Vascular endothelial growth factor-C(VEGF-C) can promote lymphangiogenesis and lymph node metastasis . The VEGF-C is found that it highly expresses and relates to lymph node metastasis in many tumor . The role of VEGF-C in nasopharyngeal remains uncleare and the correlation between VEGF-C and microlymphatic density,lymph node metastasis,prognosis of carcinoma are conflicting in the published literature. The over-expression of D2-40 has been found in many tumors and D2-40 may relates to carcinogenesis,development and metastasis of tumor.Up to now,a lot ofexperimental evidence indicated that overexpres- sion of vascular endothelial growth factor-C and D2-40 promotes angiogenesis and lymphangiogenesis . This study were designed to examine the expression of VEGF-C and VEGFR-3 protein by immunohistochemistry and observed tumor-associated angio- and lymphangiogesis using commercialized antibody D2-40 and CD34 and subsequantly quantified the LVD and MVD in human nasopharyngeal carcinoma , and evaluate their correlations each other and their correlations above parameters with clinical pathological characteristics of patients with nasopharyngeal carcinoma.Methods : The paraffin-sections of tumor tissues from 61 patients with NPC were examined for VEGF-C,VEGFR-C protein through using commercialized monoantibody D2-40 and CD34 by immunohistochemistry. Paraffin-sections of 30 mild nasopharyngitis tissues were examined as control. Then, SPSS(11.5) software was used to analyze these relationships of variaris and correlations with clinical pathologic characteristics.Results: 1.The numbers of the lymphatic vessel in peritumor area introtumor and non-tumor tissue of nasopharyngeal carcinoma were significant differently. The LVD immunostained with D2-40 in peritumor area of nasopharyngeal carcinoma was higher than that introtumor(11.01±2.99 V.S 8.83±1.78 P<0.05). There were no significances statistical among different group of sex,age and T-stages in these patients.2. Compared with control tissues, the positive stained rate of VEGF-C was 68.85% in NPC and significantly increased (10.45±4.99 V.S 7.01±2.99 P<0.05). The expression rate of VEGF-C in N stages groups was obviously different. The expression of VEGF-C had no significances statistical among different sex,age and T-stages in these patients. 3. The expression rate of VEGFR-3 was 63.64% in NPC and significantly increased compared with that in nasopharyngitis tissues. The expression rate of VEGF-C was differently in N stages(11.69±4.58 V.S 6.68±2.53 P<0.05).The expression of VEGFR-3 had no significances statistical among different sex,age and T-stages in these patients.4. The numbers of the microvessel in peritumor area,introtumor and normal tissue were statistial different,respectively.The MVD in peritumor area of nasopharyngeal carcinoma was higher than introtumer(7.68±4.11 V.S3.45±2.21 P<0.05). The tumor MVD had no statistics significances in different sex,age and T-stages .Conclusion:1.The over-expression of D2-40 is found in nasopharyn- geal carcinoma .2.The LVD in peritumor area of nasopharyngeal carcinoma was higher than that in the normal tissue and introtumer . The tumor borderline LVD was related with lymph node metastasis. These findings indicate that lymphangiogesis of peritumor area relates to lymph node metastasis.3.The MVD in tumor tissue was higher than that in the normal tissue.4.There is positive correlation between D2-40 and nasopharyngeal carcinoma MVD. There is positive correlation between VEGF-C and peritumor area LVD of nasopharyngeal carcinoma and VEGF-C and peritumor area LVD.5.The tumer-associated angio- and lymphangiogenesis in nasopharyngeal carcinoma is consistantly tendency which involves in and promotes progress of tumor.
