Time-resolved fluoroimmunoassay (TR-FIA) using lanthanide chelates as the fluorescence probes is an established immunoassay method. The most important advantage of TRFIA is that it allows easy distinction of the specific fluorescence signal of long-lived lanthanide fluorescence probe from the short-lived background signal present in most biological samples, and also obviates the problems associated with scattering light of the optical components, since the fluorescence of lanthanide fluorescence probe is long-lived with large Stokes shift and sharp emission band. Therefore the sensitivity of TR-FIA is higher than those of other immunoassay methods.This paper is a part of the State Natural Science Fund: A novel homegeneous time-resolved fluoroimmunoassay based on quantum dots labelled FRET and its application for screening for Down syndrome. Time-resolved fluoroimmunoassay was studied in my experiment.The main content in this paper:1. The technology of labling antibody with Eu3+ that is the key link in the time-resolved fluorescence immunoassay technology was studied.The factors such as reaction of pH,reaction time and temperature,concentration of protein and the ratio of BHHCT: Ab that impact on the labaling rate and protein recovery was discussed,and elected the better conditions in this paper.2. Initially established a competitive analysis of time-resolved fluorescence immune system to analyse mice IgG. It provided a limit of detection of 1.575ng/ml.3. Try to synthesis a new complexing agent BBCAP,and to study the fluorescence properties of Ab-BBCAP-Tb3+. The experiment proved that self-BBCAP meet the application requirements.
