Object:1. The optimum conditions with hot water extraction of the polysaccharides from Enteromorpha were studied.2. Studies on the effect and mechanisms of the enteromorpha's Polysaccharide on the blood lipid level,anti- oxidize and hepatic adipose infiltration in Experimental high blood lipid rats.3. Studies on the effect and mechanism of the enteromorpha's Polysaccharide on the experimental H22 tumor mouse.Methods:1. Study on the technology of extracting EP: In this procedure, water was used as solvent to extract EP. To optimize the extracting technology, single factor and orthogonal tests design were used with the four influence factors: pH,temperature,the ratio of solvent to stuff and time, each on different levels. The yield of EP was used as the optimization index.2. Study on the effect and mechanisms of EP regulating blood-fat and anti-lipid peroxidation function on rats: Adult male rats of SD grade were randomly divided into several groups according to the level of serum total Cholesterol and weight. The rats were set three doses, the low dose, the middle dose and the high dose. With the hyperlipemia model created by feeding high fat and high Cholesterol diets, the following five indexes :the serum TC, TG, LDL-C ,HDL-C and AI were determined to judge the function of blood-fat adjusting of EP on mice. The serum superoxide dismutase (SOD),glutathione peroxidase (GSH-PX) and malondialdehyde (MDA) content were determined to judge the function of anti-lipid peroxidation of EP on mice. The liver cells were observed by paraffin-sections,frozen section and optical microscope.3. Study on the effect and mechanism of EP anti-tumor function on mice: H22 cancer-bearing mice as model, EP by the different doses intervention were gavaged into H22 mice to study the anti-tumor effects and to investigate the mechanisms. The following some indexes , calculated the tumor-inhibition rate,spleen index,the serum contents of IL-2,TGF-βand VEGF,the carbon clearance index and NK cell activity were determined to judge the immune function, while its anti-tumor mechanism to investigate.Result:1. The optimal parameters of extracting EP was as follows: pH 5,the ratio of stuff to solvent was 1:80, 70℃and 2 hours. Under this condition, the yield of EP reached to 9.48%.2.The effect of EP on the adjust blood-fat and anti-lipid peroxidation function of rats: As compared to the model group ,all doses of EP could decrease serum TC, TG, and LDL-C(P<0.01).Both middle and high doses of EP can decrease AI(P<0.01). These results illustrate that the EP products are effective in adjusting the blood-fat of rats.As compared to the model group ,all doses of EP could increase serum SOD,GSH-PX(P<0.01)decrease MDA(P<0.01) . The fatty liver was prevented and cured effectively too. These results illustrate that the EP products are effective in anti-lipid peroxidation and cure fatty-liver.3. The effect of EP on the anti-tumor function of rats: Low, medium and high-dose EP can inhibit tumor growth (P <0.01), inhibition rates were16.9%,49.1% and53.1% .Low Middle and High doses of EP can increased spleen index in mice (P <0.01); high-dose EP can significantly enhance IL-2 levels (P <0.01); low, middle and high doses of EP can significant decrease in TGF-βand VEGF levels (P <0.01).Middle and high dose of the EP could significantly improve the carbon clearance index, high-dose group can significantly improve NK cell activity (P <0.01). These results illustrate that the EP products are effective in anti-tumor function and immunological regulation.Conclusions: The approach of extracting polysaccharide from enteromorpha used in this paper is a simple technology, with which can gain a considerable yield of EP was 57.2% . Moreover, the EP extracted by this approach has fine hypolipidemic, anti-lipid peroxidation and prevention of the role of fatty liver on experimental hyperlipidemia rats. At the same time, it has inhibition on experimental nude mice with tumor growth, its the possible inhibition mechanism may have something with the body IL-2, TGF-βand VEGF secretion, as well as the body of non-specific immune function, NK cell activity. Therefore, extracting EP as a new type of health care products resource has great development and utilization prospects.
