Objective: Cynoglossum lanceolatum Forsk is a common ethnic herb in southwest china.To further explore its medicinal value, with cynoglossum lanceolatum polysaccharide as theresearch object, optimize the extraction process and make protein removel and decolorization,and explore its antioxidant activity in vitro and antitumor effects.Methods: Using the method of hot water extraction, ultrasonic assisted extraction andcellulase assisted extraction method to extract cynoglossum lanceolatum polysaccharide,polysaccharide content was determined by phenol sulfuric acid method, to determine the bestprocess; using the method of Sevage and Sevage with papain method in crude polysaccharideprotein; activated carbon and hydrogen peroxide on cynoglossum lanceolatum polysaccharidedecolorization; the antioxidant ability in vitro inhibition experiments on DPPH free radical,hydroxyl radical, superoxide radical determination of polysaccharide of cynoglossumlanceolatum; through the determination of S180cell line induced mice inhibited tumor growthand physiological indexes of cynoglossum lanceolatum polysaccharide anti-tumor andenhancement the role of immunity.Results: The ultrasonic extraction method for the extraction process of cynoglossumlanceolatum polysaccharide is optimal, and the yield of polysaccharide was2.17%, theoptimum extraction conditions were: the ratio of material to liquid1:20, ultrasonic temperature60℃, extraction time50min, extraction times of2.The cynoglossum lanceolatum polysaccharide protein removal method is the best papainmethod combined with the Sevage method, the removal rate is85.7%, polysaccharide retentionrate was48.6%; the optimum method for hydrogen peroxide, the decolorization rate was93.9%,sugar retention rate is56.9%.Show cynoglossum lanceolatum polysaccharide in vitro antioxidant experiment,antioxidant ability was positively related to its concentration, on superoxide radical scavengingability on the weakest, DPPH free radical scavenging rate was the highest, the scavengingcapacity is lower than the same concentration of VC.The crude polysaccharide low concentration group, high concentration group mice S180sarcoma tumor inhibition rates were51.72%,68.29%, than the group of cyclophosphamide group; thymus index and spleen index in mice were improved, and the mice phagocytic indexand phagocytic index improved significantly.Conclusion: Ultrasound cynoglossum lanceolatum optimum method of extractingpolysaccharides. Extraction, enzyme and Sevage method combined with the best effect toprotein, hydrogen peroxide bleaching best; its scavenging free radical, has the very good effectof anti tumor and raising immunity, is a very promising medicinal...
