Background and Purpose:Nasopharygeal carcinoma is the most common one of malignant tumors in ear, nose and laryngopharynx;and it is used to putting up with lymph node and distance metastasis.In our country,especially in the south of our country,the nasopharyngeal carcinoma is hackneyed.According to WHO statistics,80%of them happenned in our country.Nuclear factor-kappa B is an important gene in adjuesting immunity and inflammation,and also is concerned with the adjuestmenrt of cell's proliferation and apoptosis.In recent,according to the research of signal translation of NF-KB,by which NF-KB is activated and other genes take part in it,we make sure that NF-KB has relationship with the tumor's occurrence,growth and metastasis.MMPs are a family of protein-enzymes,depending on znic ion,which can degrade extracellular matrix and basic membrane;then take part in malignant tumor cell's metastasis.In this study,with immunohistochemical technology,we tested the expression fo NF-KB,MMP-3 and MMP-9 in the 41 cases of NPC,and analyzed the relationship between the expression in them and the clinical information.Meterial and Method:From Jan.of 2007 to Oct.of 2008,41 cases of nasopharynx biopsy specimens were collected from patients who came to our hospital for treatment first,and didn't have other therapy before,and their diagnosis were nasopharyngeal carcinoma.The patients were 27 male patients and 14 female patients,with age ranged from 29 to 76 years old.According to UICC/AJCC classification of 2002,9 cases were in T1 stage,13 T2,10 T3,9 T4;17 cases in N0 stage,16 in N1 stage,5 in N2 stage,3 in N3 stage.We also obtained 14 nasopharynx biopsy specimens from patients,who were suffered from nasopharyngeal inflammation.All tissue samples were fixxed with 10%formaldehyde solution,then embedded in paraffin,5 micrometre thick sections were prepared for examming the expression NF-KB,MMP-3 and MMP-9 by streptaividin-peroxidase immunohistochemistry.All the figuers were analyzed statistically with SPSS 13.0 software in computure.We use x2-test to examming the difference within them,and use Spearman-test to examming the corelation of them.Result:1.Positive expression of NF-KB in nasopharyngeal carcinoma is 76.05%;positive expression of MMP-3 in nasopharyngeal carcinoma is 75.61%;and positive expression of MMP-9 in nasopharyngeal carcinoma is 76.05%too.Significant difference is between nasopharyngeal carcinoma and nasopharyngeal inflammation.2.There are significant relationship between NF-KB,MMP-3,MMP-9 and N stage. No relationship between NF-KB,MMP-3,MMP-9 and sex,age,pathological classification as well as clinical stage. 3.The expression of NF-KB showes a siginificant positive correlation with the expression of MMP-3 and MMP-9.MMP-3 also has a siginificant postive correlation with MMP-9.Conclusion:The result suggests that all of NF-KB,MMP-3 and MMP-9 are highly expressed in nasopharyngeal carcinoma cells.Their expression have relationship with lymph nodes metastasis,but do not have relationship with sex,age,pathological classification and clinical stage.Each of them has positive corelation with the others.
