Objective:To investigate the distribution of Peripheral Blood B lymphocyte Subsets(PBLS) in idiopathic thrombocytopenic purpura and evaluate the drug effect on the PBLS of ITP patients after treatment.Methods:peripheral blood CD19+cells,B1 cells in ITP Patients and healthy volulnteers were examined by a direct immunofluorescence straining technique and analyzed by 2-color flow cytometry.Results:compared with the ratios of lymphocyte subpopulations in healthy controls,CD19+,B1 cell subsets and B1/CD19+in all ITP patients significantly increased(P<0.05),After treatments,the effective cases of methylprednisolone group in ITP patients were twenty-three and refractory cases were eleven,in the effective cases CD19+,B1 cell subsets and B1/CD 19+ decreased compared with that of the ITP patients before treatments(P<0.05),At the same time,the ratios of CD19+,B1 cell subsets and B1/CD 19+ after treatments in refractory cases and rhIL-11 cases did not obviously changed(P>0.05);B1 cell subsets have inverse correlation with the peripheral Blood platelet count(r=-0.79,P<0.05 ) and have positive correlation with the platelet associated immunoglobulin(PAIgG) in idiopathic thrombocytopenic purpura(r=0.792,P<0.05).Conclusions:In relation to healthy control subject,the peripheral blood CD5+B cell(B1 cell ) subsets increased,which suggests the presence of disturbances of B lymphocyte Subsets in patients with ITP.ITP is a disease with immune function disorder of B lymphocyte Subsets,The immune function disorder can be corrected by the drug such as methylprednisolone,the examination is instructive to our diagnosis and therapy of ITP.
