ObjectiveDiscussion the vascularization VEGF for HDPPE surgical implants and supplying experimental evidence to clinical application of better.MethodsThere are 24 male adult vistar rats in all,which were divided into 2 experimental groups and 1 control group, and each group had 8 rats. 2 experimental groups was injected 200ngVEGF and 100ngVEGF topically after being implanted with HDPPE. The control group was implanted with HDPPE. We carried out general observation, hematoxylin and eosin (HE) stain and immunohistochemical stain of CD34 antigen expression 3,5,7,14 days after implantation, and march microvessel density count(MVD). MVD data was show as"(?)±S" and the use of statistical software SPSS12.0 one-way ANOVA (one-way ANOVA) and group LSD (Least significant difference)-t test analysis of the indicators to observe the differences in the average, Statistically the level of significance as a = 0.05.ResultsGeneral observation: VEGF200ng and VEGF100ng experimental groups was familiar with the control group after 3 days of implantation. HDPPE was losse combined with the surrounding tissue and a small amount of organization ingrowth. 5 days after the implantation, HDPPE was closely combined with the surrounding tissue a little closer in VEGF200ng experimental group, the ingowth organization was increased than 3 days. VEGF100ng experimental group and control group was looser than VEGF200ng experimental group and easy to dislodge. 7 days after implantation, VEGF200ng experimental group combined with the dense surrounding tissue, and the organization combined attached to the HDPPE. VEGF100ng experimental group and the control group was looser than VEGF200ng experimental group. 14 days after implantation, VEGF200ng and VEGF100ng experimental groups was similar to the control group and HDPPE combined with dense surrounding tissue, the vascular tissue fibers completely close package, it is not easy separation.HE stain: 3 days after implantation, VEGF200ng and VEGF100ng experimental group was similar to the control group. HDPPE cause the foreign body reaction, the fibroblasts and foreign body giant cells hyperplasia, the edge was infiltrated by inflammatory cell, which was mainly of Neutrophils and lymphocytes, new vessel occasional. 5 days after implantation, the control group and VEGF100ng experimental group has been a part of fibrous tissue into the central, while the central and edge of VEGF200ng experimental group both has a small amount of neovascularization. 7 days after implantation, the central of the control group and VEGF100ng experimental group has not only a large number of fibrous tissue hyperplasia, but also a small number of neovascular ingrowth, Vascular density fiber in VEGF200ng experimental group was significantly higher than the that in VEGF100ng experimental group. 14 days after implantation, all groups are visible that the central and edge of HDPPE is filled with dense fibrous tissue.Immunohistochemical stain of CD34 antigen expression and MVD: 3 days after implantation, in the edge of HDPPE the vascalar endothelial cell were positive. VEGF200ng,VEGF100ng experimental groups and the control group MVD count: 2.25±0.41,1.50±0.42,1.00±0.49. VEGF200ng group compared with VEGF100ng group and the control group, there were significant differences in statistics. 5 days and 7 days after implantation, there were more vascalar endothelial cell were positive. MVD count(5days): 3.05±0.25,2.40±0.43,1.85±0.41. MVD count (7days): 6.90±0.89,4.80±0.59,3.65±0.94. VEGF200ng experimental group compared with the VEGF100ng experimental group and the control group, there were significant differences in statistics. 14 days after implantation, a mount of vascalar endothelial cell and fibrous tissue filled in the void. MVD count: 7.65±1.07,6.90±0.89,4.40±0.43, VEGF200ng group and VEGF100ng group compared with the control group, there were significant differences in statistics.ConclusionVEGF could promote vascularize of HDPPE sugical implants after implantation and the effect of 200ng dosage VEGF was obviously.
