Objective Our previous researches have confirmed that Acanthopanax Senticosus Injection(ASI)can induce Bone Marrow Stromal Cells(BMSCs) to differentiate into neuron-like cells in vitro and these cells can express certain specific marks of neuron.This research is to observe the effect of ASI,BMSCs and their combination on ICVD.Methods BMSCs were purified and amplified by methods of adherence in vitro; Middle Cerebral Artery Occlusion (MCAO) model was copied by fishing thread, and the successful models were randomly divided into control group, ASI group, BMSCs group and combination group, adding sham group there are five groups in all.After treatment, we observed the changes of rats'neuron severity scores (NSS), morphology of brain tissue, BDNF expression level in ischemic hemisphere of brain tissue and BMSCs'survival condition. Results ASI group, BMSCs group and combination group significantly improved the rats'neurological behavior, pyramidal cell structure of ischemic cortex , increased the expression level of BDNF comparing with control group and the combination group has the most obvious approvement; The survival cells which came from transplantation in brain tissue of combination group are more than BMSCs group. Conclusions ASI and BMSCs transplantation in cmbination have synergistic effect.The mechanisms may have relation to improving microenvironment of ischemic brain tissue, promoting the survival of transplanted BMSCs and secret of BDNF'expression level.
