| Objectives: To observe the effects of fu gong zhi xue capsule on bleeding amount of uterus, coagulation time, uterus humid weight and the levels of E2, P, FSH, LH and the apoptosis of decidua tissue of incomplete termination of early pregnant rats, and discuss the role of fu gong zhi xue capsule in reducing bleeding after incomplete termination of early pregnant rats. Methods: 1. Eighty 2-month-old health Sprague-Dawley female rats and 40 2-month-old health Sprague-Dawley male rats were breed by the proportion 2:1. we examed the vaginal smear in the following morning. If we found sperma, it was the first day of gestation. Then we selected 48 pregnant SD rats, they were randomly divided into six groups including control group, model group, motherwort group, low dose fu gong zhi xue capsule group, middle dose fu gong zhi xue capsule group, high dose fu gong zhi xue capsule group. 2. The pregnant rats were treated with gastric perfusion of mifepristone(at 08:00) and misoprostol(at 18:00). The medicine abortion was successful if there was blood stain in the following morning. The rats in low dose fu gong zhi xue capsule group, middle dose fu gong zhi xue capsule group, high dose fu gong zhi xue capsule group were respectively treated with gastric perfusion of fu gong zhi xue capsule solution that amounted to 1 time, 2 times and 4 times dose of fu gong zhi xue capsule used in clinical adult patients last for five days. 3. We detected the bleeding amount of uterus and uterus humid weight of both sides, and draw blood from angular vein by glass capillary to detect coagulation time. The serum E2, P, FSH, LH were detected by radioimmune method, and the change of Fas, FasL in decidua tissue were detected by immunohistochemistry method. Results: 1. The bleeding amount of uterus in motherwort group and fu gong zhi xue Capsules compared with model group were significantly decrease(P<0.05), the high dose of fu gong zhi xue capsule had the best effect. 2. The uterus humid weight of middle and high dose of fu gong zhi xue capsules were significant lower than modle group(P<0.05), and there was no significant between motherwort group, fu gong zhi xue capsule and model group(P>0.05). 3. The coagulation time of motherwort group was significantly longer than model group(P<0.05), and it significantly shorter in middle dose of fu gong zhi xue capsule than in model group(P<0.05). 4. The level of serum E2 was significantly higher than the other groups(P<0.05). There was no significant difference between motherwort and fu gong zhi xue capsules. The level of serum P in model group was significantly higher than motherwort group and fu gong zhi xue capsules(P<0.05). 5. There were no significantly difference in The levels of FSH and LH between motherwort group and fu gong zhi xue capule groups(P>0.05). 6. The express of Fas in control group was significantly lower than the other 5 groups(P<0.05). The express of Fas in fu gong zhi xue capsule groups was significantly higer than model group and motherwort group(P<0.05), there was no significant difference between model group and motherwort group(P>0.05). The express of FasL in control group was significantly lower than the other 5 groups(P<0.05). The expresses of FasL in motherwort group and fu gong zhi xue capsule groups were significantly higher than model group(P<0.05). The expresses of FasL in fu gong zhi xue capsule groups were significantly higher than motherwort group. Conclusions: 1. The fu gong zhi xue capsule can reduce the uterus humid weight, coagulation time, and bleeding amount of uterus. It can also promote the discharge of decidua tissue in the procedure of medicine abortion. 2. Fu gong zhi xue capsule can increase the sensitive of uterus to prostaglandin to enhance the uterus's contraction force through decerase the level of serum P in order to the discharge of remain tissue and reduce bleeding. However, there were no effect on serum E2, FSH and LH. 3. Fu gong zhi xue capsule can shortern the bleeding time through adjusting the expression of Fas and FasL. |
