Part 1 Establishment of a young rat model with metabolic syndrome by giving high fat and high fat-salt diet and the study of 5α-reductase type 1 and 11β-hydroxysteroid Dehydrogenase type 1Objective:To establish young rat model with metabolic syndrome by giving high fat and high fat-salt diet,investigate the Gene expression of hepatic 5α-reductase type 1(5α-R1)and 11β-hydroxysteroid Dehydrogenase type 1(11β-HSD1)and the ratio of the two enzyme (5α-R1/11β-HSD1)in young rats with metabolic syndrome.Methods:SD young rats just Weaned,aged 3 weeks,were given r outine diet(control) and high fat diet(FC) and high fat-salt diet(FSC).Then the body weights,waist circumference,body length and blood pressure were measured 4 weeks later,an oral glucose tolerance test and insulin release test were performed,and the plasma lipids,the plasma cortisol,the amount of fat around visceral organs were measured.Hepatic 5α-R1 and 11β-HSD1 mRNA were assessed.And at the same time the Gene expression ratio of the two enzyme were calculated.Results:In the FSC group,the waist circumference,blood pressure,FPG,FINS,and serum triglycerol levels were all significantly increased compared to the control and significant insulin resistance occurred,at the same time high-density lipoprotein cholesterol significantly decreased.Hepatic 5α-R1 mRNA were significantly increased and 11β-HSD1 mRNA were significantly decreased in obese young rats with metabolic syndrome. The ratio of the Gene expression for the two enzyme were increased in obese rats with metabolic syndrome.Serum cortisol level had the significant relationship with the ratio of the Gene expression for the two enzyme(5α-R1/11β-HSD1) followed the 5α-Rlof hepatic.Serum cortisol level had the significant Negative correlation with the Gene expression for the 11β-HSD1 of hepatic.Conclusions:High liver 5α-R1 expression and low expression of 11β-HSD1may increase insulin resistance and dyslipidemia,GC metabolic disorder may be related to the occurrence and development of children MS.Targeted regulation prompted 5α-R1 and 11β-HSD1 expression and activity o may be a treatment for children MS.Part 2 Analysis of serum cortisol level and other associated factors in children metabolic syndromeObjective:To investigate the associations between metabolic syndrome(MS) and the levels of serum cortisol in obesity children.Methods:Subjects were 96 children diagnosed as obesity.Child-specific definition made by Cook was used to identify MS in these subjects.Measures included serum cortisol,OGTT and insulin release test,and serum lipid profiles,then calculated the parameters of insulin resistance HOMA-IR,following the measuring of anthropometrics measurements and blood pressure.Calculated the difference of these parameters and the correlativity of serum cortisol with other parameters.Results:Children with MS had higher serum cortisol levels,whether unadjusted or after adjusting for insulin levels.Increasing number of features of MS(nMSc) was associated with higher cortisol levels,cortisol level had the significant relationship with waist circumference followed by systolic blood pressure(SBP),diastolic blood pressure(DBP),high-density lipoprotein. The consequence of Logistic regression analysis shown WC,SBP and DBP were the important correlation factors of serum cortisol.Conclusions:Combined with insulin resistace and some other factors,cortisol may contribute to the on set and development of MS.
