| systemic lupus erythematosus is systemic autoallergic disease,which is marked by losing imbalance in immunity,including overexcitation of T and B leukomonocyte, production of multiple autoantibody,and deposition of antigen-antibody complex.All of them induce many organs are damaged.SLE happens in women in child-bearing period,the disease incidence between male and female is 1:9.The organs all over the body will be involved,SLE will threaten seriously life and healthy,which is solved urgently in clinical.The study at present,several kinds of autoimmune disease relate with losing imbalance of Th1/Th2,such as the development and turnover of SLE. So,to more realize autoimmune disease,understanding the role of T-bet/ GATA-3 in the differentiation of Th cells has significant role.Besides,Tr is found as a special kind of CD4+ T cells,which has the role of immunological regulation.Tr influences the turnover of SLE by excreting cytokines.Accordingly,to study Tr deeply has important meaning.Objective:To investigate the effect of transcription factors on the differentiation of T cells,the role of transcription factors and cytokines in the pathogenesisMethods:The expression of transcription factors T-bet and GATA-3 in PBMC from patients with SLE and healthy controls was detected by fluorescent real-time quantitative PCR,the level of IL-10 and TGF-βin serum from patients with SLE and healthy controls was detected by ELISA.Result:Fluoresent real-time quantitative PCR showed T-bet mRNA expression in patients with SLE was lower than that in controls,the differences between patient with SLE and healthy controls were significant.GATA-3 mRNA expression in patients with SLE was higher than that in controls,the differences between patients with SLE and healthy controls were significant.ELISA showed the quantity of IL-10 in serum in patients with SLE was significantly higher than that in healthy controls, and there were no significantly differences of TGF-βbetween patients and healthy controls.Conclusion:Immunity in patients with SLE loses balance between Th1/Th2, gene expression level of Th1 associated transcription factors(T-bet) are decreased,in contrast,GATA-3 that regulates Th2 cell differentiation are significantly increased. The quantity of IL-10 in patients with SLE is increased and there are not significant differences in the quantity of TGF-β. |
