| Objective:By detecting of the expression of heparanase(HPA),hypoxia inducible factor-1α(HIF-1α),CD34 protein in gastric carcinoma,this study is to investigate the relationships of HPA and HIF-1αexpression with the carcinogenesis,invasion and metastasis of gastric carcinoma,and the relativity between the expression of HPA and HIF-1αin gastric carcinoma and their effects on angiogenesis.The results may provide theoretical basis for further revealment of the invasion and metastasis of gastric carcinoma.Methods:Using tissue microarray technique and immunohistochemical SP staining method,detected the expression of HPA protein,HIF-1αprotein and CD34 protein in 80 gastric carcinoma and 12 gastric normal tissues.And the statistic method was used to analyze the expression of HPA and HIF-1αcorrelated with the expression of CD34,as well as the relationship of clinicopathological parameters.Results:1 Expression of HPA protein in various lesions of gastric epithelialThe frequency of positive expression of HPA protein in gastric carcinoma was 61.25%and showed higher expression than in normal gastric tissue(16.67%)(P<0.05).The expression of HPA wasn't correlated with age,sex,tumor magnitude or differentiation(P>0.05),but the high expression of HPA in cancer tissues was significantly correlated with fibromembranous infiltration and lymph node metastasis (P<0.05).2 Expression of HIF-1αin various lesions of gastric epithelialThe expression of HIF-1αprotein in gastric carcinoma was 53.75%and showed higher expression than in normal gastric tissue(0)(P<0.05).The expression of HIF-1αwasn't correlated with age,sex,tumor magnitude or differentiation(P>0.05),but the high expression of HIF-1αin cancer tissues was significantly correlated with fibromembranous infiltration and lymph node metastasis(P<0.05).3 Expression of HPA and angiogenesisIn gastric carcinoma,MVD with positive expression of HPA(51.21±8.30) was significantly higher than that with negative expression of HPA(32.57±7.41)(P<0.05).It indicated the expression intensity of HPA could be consistent with the value of MVD in cancer tissues.4 Expression of HIF-1αand angiogenesisIn gastric carcinoma,MVD with positive expression of HIF-1α(50.91±7.82) was significantly higher than that with negative expression of HPA(36.77±7.35)(P<0.05).It indicated the expression intensity of HIF-1αcould be consistent with the value of MVD in cancer tissues.5 The relationship between HPA and HIF-1αThere was a significantly positive correlation between the expression of HPA and HIF-1αprotein in the gastric carcinoma tissues(r_s=0.497 P<0.01).Conclusions:1 Compared with normal gastric tissues,the expression levels of HPA,HIF-1αand MVD are significantly higher in tumor tissues.It indicates that overexpression of HPA, HIF-1αand MVD are early events in gastric carcinogenesis.2 In gastric cancer,HPA and HIF-1αare related to the depth of invasion,lympnode metastasis and clinical stage.It implies that overexpression of HPA and HIF-1αproteins may play an important role in the tumorigenesis and tumor progression of gastric cancer.3 The expression intensity of HPA and HIF-1αwere consistent with the value of MVD in gastric carcinoma,it indicated that HPA and HIF-1αcould be two of the important factors in angiogenesis of gastric carcinoma.4 There was a positive correlation between the expression of HPA and HIF-1αprotein in the gastric carcinoma tissues,it indicated that there could be a positive cooperation between HPA and HIF-1αin the carcinogenesis and development of gastric carcinoma. |
