Solid-phase Time-resolved Fluorescence Immunoassay is a new immunoassay technology established,which is used to substitute radio immunity immunoassay,and have been widely applied in the basic medicine,clinic field and life science.To meet Solid-phase Time-resolved Fluorescence Immunoassay requirement.A key technique of direct solid-phase TRFIA is to design and prepare opaque solid-phase reflection immunologic reaction pool and increase S/V ratio of sample pool.It can increase absorption between polystyrene plate and detection protein,modify polystyrene plate and increase detection sensitivity.Nylon-6 is dissolved with 22mL acetic acid and 10mL m-methyl phenol is added polystyrene plate after it is soaked by pyridine:acetone(1:29) solution.Polystyrene plate smear layer is even when it is rotated at room temperature after 0.4mL pyridine is added.The labeled rate of plate and antibody is good after glutaraldehyde and hexamethylene diamond solution are connected arm by turns with the plate which is modified by alkylation. Sensitivity of it 3 times higher than routine physical adsorption method,CV<10%and stability of it is that term of validity is three months longer.Operation of the method is simple and cost of it is cheap.The method has general practical value and is applied TRFIA, RIA and ELISA.It has an important significance to study development of protein link technique.It has practical value to study conjugate of polybenzene chelate,legend and protein.
