Objective:To cultivate the periodontal ligament fibroblasts on a three-dimensional cell culture system in vitro and study the effects of cell growth factors and pathogenic metabolites on the growth of those cells in regular cell culture and three-dimensional cell culture conditions.Methods:Human periodontal ligament fibroblasts(HPDLFs) were cultured by tissue explants. The primary cells were used for experiments after 4 passages.The cells were seeded on a three-dimensional cell culture system and regular cell culture plates respectively.The characteristics of those cells were studied under microscope.The cell growth under those two conditions were examined and evaluated by using MTT assay.In addition,the basic fibroblast growth factor and pathogenic metabolites(lipopolysaccharide) at different concentrations were added in those cell cultures at designated time points.Results:The cell growth on the three-dimensional cell culture is similar with the two-dimensional cell culture without stimuli.But when adding 10ng/ml basic fibroblast growth factors,the cell growth was significantly higher than control.However,the cell growth was inhibited when adding 100ng/ml lipopolysaccharide in those three-dimensional cell cultures.Conclusion:The cell growth on the three-dimensional cell culture is similar with the two-dimensional cell culture in this experimental condition.The low concentration of the basic fibroblast growth factor could increase the growth of HPDLFs.The low concentration of lipopolysaccharide could inhibit the growth of HPDLFs.
