Objective:To investigate the correlation between the expression of FHIT,PTEN and Ki-67 and their roles in the genesis and development of lung cancer in Xuan Wei region.Method:Using immunohistochemical staining and in situ hybridization,40 cases of lung cancer in Xuan Wei region and peri-cancer tissue were abserved for FHIT,PTEN, Ki-67 proteins and FHITmRNA with Dig-labeled probe.Tissue microarray technique were used to make tissue blocks.Results:(1)The positive expression rate FHIT was 35.00%(14/40).The rate of expression of FHIT proteins in tumor tissues was significantly lower than that in non-tumor tissues(x~2=38.52,P<0.05).There was a significant difference between squamous cell carcinoma 61.54%(8/13) and adenocarcinoma 22.22%(6/27)(P=0. 01).FHIT wassignificantly correlated with the histology,the degree of differentiation of tumor and sex(P<0.05).There was no relationship between FHIT expression and age,tumor size,clinical staging,lymph node metastasis(P>0.05)(2)Lung cancer in Xuan Wei region,the positive expression rate of PTEN protein were 37.50%(15/40). The positive expression of PTEN protein were detected in a significantly lower proportion in lung cancer in Xuan Wei region than in sampeas peri-cancer of lung cancer in Xuan Wei region,(x~2=36.36,P<0.05 );There was no relationship between FHIT expression and the histology,sex,age,tumor size(P>0.05);otherwise,there was closely relationship between FHIT expression and the degree of differentiation of tumor and clinical staging,lymph node metastasis(P<0.05).(3) The positive expression rate Ki-67 was 70.00%(28/40) lung cancer in Xuan Wei region.The rate of expression of Ki-67 proteins in tumor tissues was significantly higher than that in non-tumor tissues(x~2=43.08,P<0.05).Ki-67 was significantly correlated with the degree of differentiation of tumor(x~2=6.81 P=0.01).There was no relationship between FHIT expression and the histology,age,sex,tumor size,clinical staging,lymph node metastasis(P>0.05)(4) Expression of FHIT was not correlated with express of PTEN(r=0.63,P=0.37); Expression of FHIT was negative correlated with expression of Ki-67(r=-0.95, P=0.05);Expression of PTEN was also negative correlated with expression of Ki-67(r=-0.80,P=0.02).(5) IHC positive in 14 cases,CISH has 5 cases of gene amplification,amplification rate of 35.71%(5/14);IHC negative 26 cases,CISH gene amplification for the 13 cases,amplification rate of 50%(13/26).Lung cancer in Xuan Wei region FHIT-cDNA lower than in adjacent normal lung tissue,the difference was significant (P<0.01).Conclusion:(1) FHIT gene was low expression in lung cancer,particularly in adenocarcinoma cells in Xuan Wei region,but Ki-67 protein showed high expression in the tumor cells which show low expression of FHIT,suggesting that FHIT gene may play an important role in lung cancer in Xuan Wei region caused by coal-fired.(2) PTEN gene loss or mutation may play an important role in the progress of lung cancer.(3) FHIT,PTEN and Ki-67 protein were detected in the lung cancer in Xuan Wei region to help Judging prognosis.
