Objective: To investigate the expression of suppressor gene FHIT (fragile histidine triad) , PTEN (phosphatase and tensin homology deleted on chromosome ten) and its significance in non-small cell lung cancer (NSCLC). Methods: (1) 34 non-small cell lung cancer tissue samples and 14 normal lung tissue samples in benign pulmonary diseases were selected randomly in Zhuang People.(2) The FHIT mRNA and PTEN mRNA were amplified by reverse transcription polymerase chain reaction ( RT–PCR ) in 34 cases of NSCLC and 14 cases of benign pulmonary diseases lung tissue .Results: (1) The loss of FHIT expression of normal lung tissues, non-small lung cancer was 0, 67.6%, respectively. Loss of FHIT expression in NSCLC was significantly higher normal lung tissue (P<0.001). The loss of FHIT expression in NSCLC was related to smoking(P<0.05), but not to age, sex, TNM stages, histologicol styles, degree of cell differentiation tumor and lymph node metastsis(P>0.05).(2) The rate of loss of PTEN expression in non-small cell lung cancer was 64.7%(22/34) which was significantly higher than that in tissues of in normal lung tissues 7.2%(1/14) (P<0.001). The loss of PTEN gene was associated with pathological histologicol styles in lung cancer patients (P<0.05), but not with age, sex, smoking, degree of cell differentiation tumor,TNM stages, Lymph node metastasis(P>0.05).(3) Acurate rate of detection NSCLC was to 67.6%, when FHIT and PTEN combine. But the difference wasn`t statistically sigfinicant between FHIT and PTEN in NSCLC(P>0.05).Conclusion:(1) The loss of FHIT in Zhuang People NSCLC was more than that in normal lung tissue, which illustrated that the loss of FHIT was a frequent, and the dysfunction of FHIT played an important role in the process of lung carcinoenesis.(2) Smoking may result in increase in loss of FHIT and incidence of NSCLC.(3) There was a higher incidence of negative expression of PTEN mRNA in tissue of Zhuang People NSCLC, which indicates that PTEN gene inactivation may play an important role in the genesis and development of NSCLC.(4) In the process of lung carcinogenesis in Guangxi Zhuang People, FHIT and PTEN are relatively independent of their case, but both united detection, can improve lung cancer detection sensitivity and negative predictive value.
