Objective:Under the instruction of theory of Chinese Medicine processed method,we investigated the fingerprint of Chebule.Besides,after an assessment on the antibacterial ability and antioxidation of Chebule we can supply experiment foundation for clinic use with different processed Chebule.We also have made a standardize study of the quality standards of Chebule formula granule,which will provide scientific criterion and referable demonstration to promote the quality standard of Chebule formula granule.Methods:Using gallic acid as a comparision,we measure the content of gallic acid and carry on the activity of solubility lixiviating for different processed Chebule.Using TLC and HPLC methods,we studied the fingerprint of different processed Chebule.We also find out the influence of different processed Chebule on the content of T-AOC,SOD,MDA and MAO-B in rat blood serum,on the content of LF and MAO-B in rat liver and on the content of T-AOC and SOD in D-galactose elder rat blood serum,which greatly support the clinic experiment.On the basis of the grained Chebule we already have,we promote and optimize the quality standard of Chebule formula granule.Results:Quality Standard Research for Different Processed Chebule:The result of our measurement on the content in different processed chebule shows that the content of gallic acid for Chebule after fry,burn and bran is higher than normal, which can work as experiment support for the theory of increasing acerbity and stopping diarrhea.We also plot the result that the shape of Chebule is related to the content of gallic acid inside it and the activity of water-solubility lixiviating and wine-solubility lixiviating accelerate when the Chebule was processed.Fingerprint Research Result: TLC fingerprint result shows that there is not much difference in the four processed samples except the content difference.And when using above mentioned method to analyze the content of multi-phenolic acid,it is better to twist the folium twice using acetic ether-formic acid(3:1) impregnant.When we perform Folium chromatogram analysis on triterpene we find that using developing solvent of different polarity the resolution and definition will be greatly increased.The study of HPLC fingerprint shows that the four samples have much similarity,but the quantity in the chromatogram apex is different.In sectionⅠ,No.1~#,2~#,3~# apex all appeared,but wheat branned Chebule do not have No 4~#.And the arrange of the four apex distribution is No.1~#>>2~#<3~#(gallic acid)>4~#.The quantity of fried Chebule,wheat branned Chebule and sand burned Chebule is bigger than Chebule for No 3~# apex,which changes the quantity ratio of No 1~# and 3~#.In sectionⅡ,the quantity of No 5~#,6~#and 10~# show a tendency of increase,but it is not the same for the No 10~# apex in branned Chebule, And the continuous show up of No 8~#,9~# and 10~# apex consists of the main character of Chebule.The No 5~#,6~# and 7~# apex of fried Chebule,sand burned Chebule and wheat branned Chebule is a little lower than Chebule.A sharp single apex appeared for wheat branned Chebule,which makes it different from others.The sectionⅢcontains No 11~#-17~# apex,in which No 11~# and 13~# are small and No 14~# and 16~# are the highest peak.The main character of sectionⅢis the pronouncing difference between the highest apex and others. The quantity of No 14~# and 16~# apex of different processed Chebule is different.The No.16~# apex in the processed Chebule increase in quantity,in which the fried Chebule increase the most.The Study of primary pharmacodynamics:Our experiment of the influence of processed Chebule on the anti-oxidation effect of rat shows that processed Chebule has significant anti-oxidation effect,in which the performance of fried and branned one is better.The study on the anti-oxidation effect of rat shows that Chebule,the sand fried and the wheat branned one can increase the content of T-AOC in rat serum(P<0.01 or P<0.05).The branned Chebule can increase the content of T-AOC in rat serum than Chebule.Compared with Chebule the livability is not much difference.Result of Study on the Quality Standard:On the basis of the existing quality standard of Chebule formula granule,by the methods of TLC,HPLC and so on,we made systematic study on the quality standard items,including Chebule formula granules" appellation,source,preparation,character, identification,assaying,funtion and indications,usage and dosage,notice,package,storage and shelflife,etc.We formulated the quality Standards of Chebule formula granule,and wrote the explanation of quality Standards,made further improvement of the quality control level of the chinese traditional medicine formula granula.Conclusions:Directed by the theory of T.C.M.,we investigated the primary pharmacodynamics and fingerprint of Chebule.Set up the database of TLC and HPLC fingerprint for different processed Chebule.Experimentally studied the antibacterial and anti-oxidation of processed Chebule,which can be regard as clinic support.We improve the existing quality standard and add the item Of measuring the content.Our work will be helpful for the standardization of the existing standard of Chinese formula granules.
