The quality standard of traditional Chinese medicine Hemsleya omeiensis is imperfect,and its taste is extremely bitter.It isn't accepted by patients easily.Cutting into slices and daying is its traditional processing method,but it can't c hange its poor acceptability.Based on TCM theory,the research processed Hem sleya omeiensis with glycyhiza jiuice in order to reduce Hemsleya omeiensis's bitterness,increase its function and decrease its toxicity by glycyhiza's sweet flavour and its function of moderating the property of herbs.Hemsleya omeiensis is research object.Its quality standard and pharmacological actionl were studied.ObjectiveTo study on row Hemsleya omeiensis product and its processed product's quality control with fingerprint,stability,anti-inflammatory and analgesic effects and toxicity,espected to improve the quality standard of Hemsleya omeiensis and provide rationality and scientificity of itr processed product with glycyhiza.Methods:1.HPLC was employed to determine the content of hemslecin A.Taking content of hemslecin A as index,the orthogonal test and the central composite design-response surface methodology(CCD-RSM)were adopted to optimize the processing technology of Hemsleya omeiensis processed by licorice juice.2.Microscopy,HPLC,TLC,etc were applicated to improve the quality standard of Hemsleya omeiensis.3.HPLC-DAD was used to establish HPLC fingerprint of Hemsleya omeiensis based on10 batches.4.Stability study was completed by drug comprehensive stability test chamber.5.The study also researched the inflammatory effects of row Hemsleya omeiensis and its processed products by using the acute inflammatory models ofxylene-induced ear edema and egg white-induced paw edema;Meanwhile,the analgesic effects of were observed by hot plate and acetic acid writhing test.6.According to the classic acute toxicity(the majority of the lethal dose LD50,the maximum tolerance MTD,the maximum dosage MLD),the study on the toxicity of Hemsleya omeiensis was conducted before and after the preparation.Results:1.The optimum processing technology of Hemsleya omeiensis by the orthogonal test was covered moistening time of 7 h,drying-time of 12 h,and processing temperature at 80 ?.The optimum processing technology by CCD-RSM was covered moistening time of 7.48—8.56 h,drying-time of 12.06—13.12 h,and processing temperature at 80 ?.2.The study of the quality is improved,The identification condition of thin layer was optimized,and the method of determining the content of hemslecin A and the fingerprint spectrum of 10 batches from differents areas were established.3.The row and processed Hemsleya omeiensis powder the powder packing of the powder was sealed package,specification 1g/ bag,store in cool and dry place,valid for 2 years.the specification 1g/ bag,store in a cool anddry place,valid for 2 years.4.Compared with the model group,the low-dose and high-dose groups of row product and processed product both had inhibition effects on ear edema of mice(P<0.01)and paw edema of rats(P<0.05~0.01).And,they also were effective at increasing pain threshold in the hot plate test(P<0.05~0.01)and reducing the number of writhing reaction in acetic acid writhing test(P<0.05~0.01).In addition,there were significant differences betweenthe high-dose groups of processed Hemsleya omeiensis L.T.Shen et W.J.Chang and high-dose groups of its raw product in all indexes(P<0.05~0.01).Though the low-dose groups didn't all show significant differences,the low-dose of processed product had better treatment effect.5.The row Hemsleya omeiensis' s MTDvalue was 20.0 g· kg-1 · d-1,and the processed Hemsleya omeiensis' s MTD value was greater than 20.0 g· kg-1·d-1.ConclusionsThe study studyed the processing technology of Hemsleya omeiensis processed by licorice juice and proved its quality standard(character,TLC,and content determination and fingerprint,etc.),stability test,anti-inflammatory and analgesic effects which provide reference for the establishment of the quality standard of Hemsleya omeiensis and basis for the rationality and scientificity of the processing Hemsleya omeiensis with glycyhiza.
