Objective:The purpose of this study was to investigate whether or not adenosine is effective in reducing lung ischemia-reperfusion injury in rats.Methods:80 rats were randomly divided into four groups:the control group(group C,n=20);the ischemia/reperfusion group(group I/R, n=20);the adenosine group(group A,n=20);the adenosine and Dipyridamole group(group AD).Group C:received sternotomy only and no ischemia;Group I/R:the left lungs of rats were rendered ischemia by ligating the left pulmonary hili for 60 minutes followed by 120 minutes reperfusion.Group A:the left lungs were rendered ischemia for 60 minutes followed by 120 minutes reperfusion,adenosine 0.1mg·kg-1·min-1 was administered intravenously 30 minutes prior to ischemia through femoral vein. Group AD: pretreated the ischemic-reperfusion model with dipyridamole(0.25mg·kg-1 ) bid before the operation the left lungs were rendered ischemia for 60 minutes followed by 120 minutes reperfusion,adenosine 0.1mg·kg-1·min-1 was administered intravenously 30 minutes prior to ischemia through femoral vein. The lung tissues were collected after the end of the operation.The W/D,MDA,MPO,SOD,Apoptotic index were measured.Results:(1) lung tissues of group I/R were injured obviously.(2) W/D,MDA,MPO,AI were much lower in group A and group AD than those in group IR(all P<0.05);The SOD level was significantly higher in group I/R than this in group A and group AD P<0.05).(3)The damage in lung tissues of group I/R were greater than that of group A and group AD under optical microscopy and electronmicroscope.Conclusions:(1)Adenosine can attenuate lung ischemia reperfusion injury in rats.(2) Adenosine may have protective effect on the lung ischemia-reperfusion injury through inhibition of polymorphonuclear neutrophils aggregation and activation,reduction the injure of oxyradical and reduction of lung tissue edema.(3)Dipyridamole which is a inhibitor of phosphoesterase can improve microcirculation and rasise the content of adenosine in tissues to attenuate lung ischemia-reperfusion injury.
