Objective: To observe the different groups of rats myocardial adenosine content changes in the organization,and the electroacupuncture(EA)preconditioning on myocardial ischemia-reperfusion injury(MIRI)the effect of adenosine in the myocardial tissue of rats,to explore the curative pretreatment of MIRI rat myocardial protection mechanism of prevention.Methods: 40 SPF male SD rats were randomly divided into five groups,with 8 in each group,and were treated separately,once daily for 7 days.Rats of the control group were only banded for 30 minutes.Rats of the sham operation group were underpassed through the artery with a thread,But didn't ligature the coronary artery,the other three groups were made MIRI models.the model groups only were made models without EA;the other two acupuncture preconditioning groups were respectively electricacupunctured at neiguan and hegu points for 30 minutes,then ligaturing the left anterior descending branch of coronary artery for 40 minutes,go on to recovering the perfusion of coronary artery for 60 minutes.In order to make the model successfully,the experiment should be monitored by electrocardiogram in the entire process.The left ventricular myocardium of the lower segment of anterior descending branch of the left coronary artery were collected in 5 groups of rats.The changes of myocardial pathological morphology were detected by HE staining and transmission electron microscopy,and the content of ATP,ADP,AMP in myocardial tissue was determined by HPLC.Results:1.Compared with the control group and the sham operation group,the model group suffered severe myocardial injury.Compared with the model group,the myocardial injury was mild in the EA PC6 and EA LI4 group.the myocardial injury in the EA LI4 group returns to normal.2.the sham operation group and the model group,the contents of adenosine were decreased in comparison with the control group(p < 0.05,p < 0.05).while the contents of adenosine in the model group were also decreased in comparison with the sham operation group.However,Compared with the model group,the contents of adenosine in the EA PC6 group were increased(p < 0.05),and the contents of adenosine in the EA LI4 group were significantly increased(p < 0.01).In addition,Compared with the EA PC6 groups,the contents of adenosine in the EA LI4 group were higher.But,the contents of ATP,ADP and AMP in the EA LI4 group were obviously higher than the EA PC6 group(p < 0.01,p < 0.01,p < 0.01).Conclusion:EA PC6 and LI4 may reduce the pathological injury of myocardium and increase of adenosine in myocardial tissue,they have preventive and protective effects on MIRI rats.What's more,the preventive and protective effect of EA LI4 is better than EA PC6.
