Background: Diabetic nephropathy is one of the most common and severe chronic complications of diabetes mellitus . It affects quality of life and survive rate of the sufferers. The main pathological alteration is the accumulation of extracellular matrix aggregation on renal tubular and renal glomerulus.Accordingly,it can result in glomerular sclerosis and renal interstitial fibrosis.The prevention and cure to progression of kidney fibrosis has already been the focus of diabetic nephropathy research. Adiponectin ,an adipoctye -derived protein, could defend against diabetes mellitus . Numerous studies have demonstrated that adiponectin has multiple functions, such as, anti-inflammatory, antiatheroscierotic and anti-fibrosis. Hence, Adiponectin is expected to be one of the new methods for the treatment of diabetic nephropathy.Objective: To investigate the effects of adiponectin (ADPN) on cell proliferation and the expressions of transforming growth factor-β1(TGF-β1) and intercellular adhesion molecule-1(ICAM-1) in human renal tubular epithelial cell cultured with high concentration of D-glucose and to illuminate the potential protection of ADPN on the kidney.Methods:①The human renal tubular epithelial cells were cultured with 5.5 mmol/L glucose (control group) , 25.0 mmol/L glucose (high glucose group) , 25.0 mmol/L glucose and concentration of 2.5μg/ml adiponectin (ADPN group), respectively. After culture for 24 h,48 h,96 h, the proliferation activity of human renal tubular epithelial cell was observed by MTT .②The human renal tubular epithelial cells were cultured with 5.5 mmol/L glucose (control group) , 25.0 mmol/L glucose (high glucose group) , 25.0 mmol/L glucose and different concentration of adiponectin: 2.5, 5.0, 7.5, 10.0, 15.0, 20.0μg/ml , ADPN group). The expressions of TGF-β1 and ICAM-1 were measured by enzyme-linked immunosorbent assay(ELISA) after culture for 48 h.Results: The proliferation of human renal tubular epithelial cells was inhibited in high glucose group and promoted time-dependently in low concentration adiponectin(2.5μg/ml)group. The expressions of TGF-β1 and ICAM-1 increased in high glucose group.The expressions of TGF-β1 and ICAM-1 were suppressed dose-dependently in moderate and high concentration adiponectin group(10.0,15.0,20.0μg/ml). Conclusion: The high density D-glucose could inhibit the proliferation activity of human renal tubular epithelial cells and promote the expressions of TGF-β1 and ICAM-1. The low concentration adiponectin(2.5μg/ml)could promote the proliferation activity of human renal tubular epithelial cells . Moderate and high concentration adiponectin(10.0,15.0,20.0μg/ml) can inhibit the expressions of TGF-β1 and ICAM-1 in human renal tubular epithelial cells .The result prompts: adiponectin maybe protect the diabetic kidney by decreasing the expressions of TGF-β1 and ICAM-1.
