Objective:To study the possible role of heat shock protein 70 monoclonal antibody (HSP70) on cultured human pancreatic cancer cell line Bxpc-3 growth inhibition and apoptosis and to clarify the heat shock protein 70 monoclonal antibody at the role of pancreatic disease and the possible mechanism.Methods:The human pancreatic cancer Bxpc-3 cells were cultured in vitro and were cultured in different medium conditions. Bxpc-3 cells were randomly divided into a blank control group and an experimental group.The experimental group was separately added with different concentrations of heat shock protein 70 monoclonal antibody with the final concentrations of 10μl / ml, 20μl/ml and 40μl/ml. Then Bxpc-3 cells were treated in 24h,48h with the application of thiazole blue (MTT) colorimetric assay to test the proliferation activity of Bxpc-3 cell.Results:The MTT results showed that the effect of different concentrations of heat shock protein 70 monoclonal antibody on Bxpc-3 cells has a statistical significance in absorbance (A) between the experimental group and the control group (P <0.05) after 24h and 48h. The inhibition rate of high concentrations group was higher than the low concentration group and the inhibition rate in 48h was higher than in 24h. Flow cytometry analysis showed that Bxpc-3 pancreatic cancer cell apoptosis in 48h by HSP70 monoclonal antibody and with the concentration of monoclonal antibody.increasing,the apoptosis rate.was also increased.The difference has a statistical.significance between the different concentration of groups.(P <0.05).Conclusion:1.Heat shock protein 70 monoclonal antibody can inhibit human pancreatic cancer Bxpc-3 cell proliferation in vitro depending on the concentration and the time.2.Heat shock protein 70 monoclonal antibody can induce human pancreatic cancer Bxpc-3 cell apoptosis in vitro and apoptosis happens with the monoclonal antibody concentration of positive correlation.
