Objective To explore the expression and their clinical significance of Syk(spleen tyrosine kinase)gene and its methylation in cervical carcinoma.Methods Using RT-PCR technique,specimens from 60 cervical cancer patients,50 cervical intraepithelial neoplasia(CIN) patients,20 normal cervical tissues were detected for their expression of the Syk gene.Meanwhile,Methylation-specific PCR was used to detect methylation of Syk promoter region.Results The expression of Syk gene were detected in 20 normal cervical tissues and 18 CINI samples,17 out of 32 CINⅡ~Ⅲsamples were found the expression of Syk mRNA,and 21 out of 60 cervical carcinoma samples were found the expression of SYK mRNA,Syk expression in cervical carcinoma was lower than that in normal cervical tissues and CIN samples(P<0.05),at the same time,the expression of Syk was higher in CINI than that in CINⅡ~Ⅲ(t=6.65,P<0.05).In 13 cases with lymph node metastasis,1 were found the expression of Syk mRNA;In 47 cases with no lymph node metastasis,20 were found the expression of Syk mRNA,a significant difference was noted between two groups(P<0.05).34 out of 60 cervical carcinoma samples were detected methylation of Syk gene in promoter region,normal tissues and CIN tissues were not detected methylation of Syk gene in promoter region.In 13 cases with lymph node metastasis,11 were found the methylation of Syk gene;The rate of Syk promoter in cervical carcinoma was higher than that of in normal tissues and CIN tissues(X~2 =13.37,P<0.05;X~2 =7.53,P<0.05).The expression of Syk gene and methylation of Syk gene in promoter region in cervical carcinoma were significant associated with the lymph node metastasis (P<0.05).While there were no significant relationship with the age/tumor's size/clinical stage/histological size and the pathological types of the tumor(P>0.05).There was a significance relationship between methylation of Syk gene and the expression of Syk gene(P<0.05).Conclusion Hypermethylation leads to lose of the Syk gene expression in human cervical carcinoma。Methylation of Syk promoter may be correlated to oncogenesis, metastasis of cervical carcinoma.
