Objective:To investigate the expression of Syk gene in human carcinoma of endometrium and to investigate the correlation between promoter region hypermethylation of Syk gene and oncogenesis,metastasis in carcinoma of endometrium.Methods:Using RT-PCR technique,specimens from 52 carcinoma of endometrium patients,30 atypical hyperplasia endometrium,28 complex hyperplasia endometrium,30 simple hyperplasia endometrium,and 40 normal endometrium were detected for their expression of syk gene,meanwhile,nest duplicated methylation -specific PCR was used to detect methylation of syk promoter region.Results:The positive expression of Syk gene is gradually increased in endometrial carcinoma tissues,endometrial atypical hyperplasia tissues,endometrial complex hyperplasia and endometrial simple hyperplasia tissues,and normal endometrial tissues,The rate of methylation of Syk gene is gradually depressed (P<0.05).However,there is no difference in endometrial complex hyperplasia and endometrial simple hyperplasia tissues,and normal endometrial tissues(P>0.05).The positive expression Syk gene and the rate of methylation of Syk gene is correlative with the clinical stage of endometrial carcinoma(χ~2=4.65,P<0.01;χ~2=4.33, P<0.05),lymph node metastasis(χ~2=10.68,P<0.01;χ~2=5.86,P<0.05),and depth of infiltrating muscular layer(χ~2=5.82,P<0.05;χ~2=3.90,P<0.05),A significant difference was noted between the two groups,but there is not related to histological grade of endometrial carcinoma(χ~2=0.03,P>0.05;χ~2=0.94,P>0.05).Syk gene hypermethylation is related to the lose of the syk gene expression in human carcinoma of endometrium(P<0.05).Conclusion:Hypermethylation leads to the lose of the syk gene expression in human carcinoma of endometrium.Methylation of Syk may play an important role in tumor infiltration and matastasis in carcinoma of endometrium.
