Investigate The Distribution Feature Of Bc16 Gene Isolation Of DLBCL In Guangxi

Object:Investigate the feature of bcl6 gene isolation with the relationship among the type of amynology,age,and the index of tumor increment in DLBCL, research the role of bcl6 gene isolation in the development of DLBCL,and the possible effect on prognosis.Method:①Selected 220 DLBCL samples by HE and IHC,and then make these samples into tissue microarrays.Use ISH to detect CD10,BCL6,MUM1 dividing DLBCL into two subgroups:germinal center(GCB) type and non-germinal center(non-GCB) type.②Use IHC to detect the expression of Ki67 in DLBCL.③Use FISH to detect bcl6 gene isolation in the two subgroups.④Use statistic software to analyze the relationship of bcl6 gene isolation with BCL6 protein expression,and value-added degree of tumor cells in different subgroups and age.Result:(1)According to the expression of CD10,BCL6,MUM1,in 220 DLBCL samples,72 cases(32.73%) are GCB type,148 cases(67.27%) are non-GCB.24 cases(25.53%) of 94 nodal cases are GCB type;48 cases(38.10%) are GCB type.Among 113 DLBCL cases that older than 50,31 cases(27.43%) are GCB type;among 107 cases of younger than 50,41 cases(38.32%) are GCB type.(2) In 220 DLBCL,83 cases(37.72%) have bcl6 gene isolation and 63/81 cases(75.90%) are in the non-GCB group,the bcl6 gene isolation has statistical differences in two group(P=0.034,＜0.05),and extra-nodal cases higher than that of nodal cases.(3) 94 cases(42.73%) have BCL6 protein expression in all cases,GCB group has 47 cases(65.28%),the expression of BCL6 has statistical differences between two groups(p=0.000,＜0.05) and positive expression of BCL6 is higher in GCB group.(4) 127 cases(57.27%) have high expression of Ki67 in all cases,GCB group has 36 cases(50.00%) high expression of Ki67,non-GCB group has 91 cases(61.49%) high expression of Ki67,the expression of Ki67 is higher in group non-GCB but there is no statistical differences between two groups.(5) In all the cases,the correlation of bcl6 gene isolation with bcl-6 protein expression and Ki67 protein expression have no statistical differences. The higher tendency of correlation maybe occur in the group of non-GCB that older than 50.Conclusion:The research reveal there were no disposition difference between age and sexuality among DLBCL in Guangxi,but more cases were non-GCB.Bcl6 gene isolation is common in DLBCL in Guangxi,about 37.72%, much more in non-GCB group,and the patients who have bcl6 gene isolation are always have high expression of Ki67 and more older.The bcl6 gene isolation maybe have correlation with the morbidity of non-GCB,we should put more emphases on etiology and prognosis.
