Establishment Of A Human Nasopharyngeal Carcinoma Drug-resistant Cell Line Tw03/DDP And The Relationship Between Drug-resistance And Lung Resistance-related Protein

OBJECTIVE:This study was designed to establish a Human Nasopharyngeal Carcinoma Drug-Resistant cell line and to investigate the relationship between drug-resistance and lung resistance-related protein.METHODS:A cisplatin-resistant human nasopharyngeal carcinoma cell line,Tw03/DDP,was induced by continuously esposing and gradually increasing dose of cosplatin.The resistant indes was estimated by MTT assay.The cell cycle was estimated by flow cytometry.Cell growth curve,doubling time and cellular appearance were measured.The expression of LRP was measured by immunocytochemisty,western blotting and RT-PCR.RESULTS:The resistant index of Tw03/DDP to cisplatin,5-fluorouracil Adriamycin,cytrarabine and Vincristine were 40.85,5.65,26.68,1.60,28.19 respectively.And the differences between Tw03/DDP and Tw03 have conspocuous statistical significances.The proliferation of Tw03/DDP got faster;the doubling time is decurtated;and the cell number of G2/M-phase increased while G1/M-phase deceased compared with Tw03.The LRP mRNA and LRP were both up-regulation in Tw03/DDP compared to Tw03 which were detected by immunocytochemisty,western blotting and RT-PCR.CONCLUSIONS:The cisplatin-resistant cell line Tw03/DDP was established which showed a typical resistant phenotype to anti-cancer drugs.The drug-resistance about Tw03/DDP has correlate to the up-regulation of LRP.
