CDH4 is a member of the cadherin family encoding for R-cadherin.It is reported that CDH4 may act as a tumor suppressor gene in human gastrointestinal tumors and proteintially be used as an early diagnostic marker for gastointestinal tumorigenesis. In the previous studies we have investigated that the expression of CDH4 was silenced in NPC cell lines CEN1,CNE2, TW03,C666-1,and express in the normal nasopharyngeal epithelia cell line NP69.Promoter methylation of CDH4 could be detected in the all the CDH4-silenced cell lines of the NPC cell lines and 90%tumors,but not on any of the normal epithelia.Loss of CDH4 can be greatly restored by the methytransferase inhibitor 5-aza-dC in NPC cell lines.Ectopic expression of CDH4 in CDH4-silenced and-methylated NPC cell line HONE1 shows that CDH4 could inhibit cell colony formation and cell migration, can improve the cell-cell attachment, which provided further evidence that CH4 is a candidate tumor suppressor gene.In conclusion,CDH4 is possible to act as a candidate tumor suppressor gene(TSG) in NPC...
