Experimental Study Of Angiogenesis Effects In Rats With Focal Cerebral Ischemia And Reperfusion Injury And Its Mechanisms By Hexueshengluo Method

Objective: To study whether Hexueshengluo Method has the effect of Therapeutic Angiogenesis after focal cerebral ischemia and reperfusion rats. And we have studied the contents of VEGF at blood serum level in rats to investigate the mechanisms of Hexueshengluo Method.Methods: The Middle Cerebral Artery Occlusion(MCAO) was ligated to establish model of focal cerebral ischemia in Wistar rats, and then they were randomly divided into 3 groups, and treated by different therapies. These groups were group of HXSLF Method, the model group, the sham operated group. During all the experiments all rats were fed ordinary food by specific person and get their food and drink freely. All of the operated rats were treated in 24 hours after their operation. The doses of their medicine were equal as adult human. All of their doses in different groups were like this: Hxsl group, 8ml/kg/d; the model group and sham group were treated with normal saline at the equal volume. We observed their general status, the score of neurological deficit, micro-vascular density(MVD) in the edge of infarcted section to decide whether Hexueshengluo Method could induce angiogenesis and investigated the contents of VEGF at blood serum level to decide whether the method could promote the expression of the key factor angiogenesis effectively.Results:1 General condition comparison: HXSL method could improve the general condition of focal cerebral ischemia rats obviously; It could obviously improve their condition, rats'activity, superficial glossiness, diet and so on.2 The scores of neurological deficit: The scores deficit in HXSL group were lower than that in the model group (p<0.01).3 Blood serum VEGF content: At different time point, the expression of blood serum VEGF content in HXSL group were higher than that in sham group and in model group.4 The microvessel density(MVD): MVD in HXSL group were higher than that in sham group and in model group.Conclusions: The prescription of HXSL method can improve the focal cerebral ischemia rats'general condition, reduce the scores of neurological deficit, and help to improve the establishment of collateral circulation, induce angiogenesis. Its mechanisms possibly stated in improving VEGF level. VEGF is the key factor of angiogenesis , which can strongly induce angiogenesis when lack of oxygen and blood.Our research convinced Hexueshengluo Method's angiogenesis effect and provided theoretical and experimental basic for clinical use of the method to cure Cerebral Infarction.
