Objective: At present, the chemotherapy is still one of the main means curing the cancer effect, a series of toxic side effects can be led by chemotherapy since chemotherapy medicine has no-choice to destroying normal tissue cells and tumour cells. Among them, myelosuppression was more common. Chemotherapy wasn't implemented on schedule when it was serious. Which results in chemotherapy failure. How to reduce myelosuppression is the key to the success of chemotherapy from some saying. So it is very important for seeking effective methods of controling myelosuppression. At present, these are some ways that modern medicine cures the myelosuppression which is caused by chemotherapy, for example, leukocyte elevating oral drug, such as Leucogen, betyl alcohol, vitamin B4 etc, the curative effect of them are not good relatively; the other way is that blood components transfusion or hematopoietic stem cell transplantation, but both of them are not easy to repeatedly use and extensively popularize. These are current new types of leukocyte elevating drug such as rHuEPO(ESPO, Epogen etc); rHG-CSF(Gran,Granocyte, rHG-CSF,etc),rHuGM-CSF(Leucomax),Thrombopoietin,interleukin-11(rhIL-11, juheli, mega etc). All these drugs have definite curative effect, but they are high in price. We make comprehensive reference to the reports that traditional chinese medicine therapy on myelosuppression in the recent years, we find that some people use the way of invigorating qi and blood, some people use the way of strengthening spleen and reinforcing kindney to replenish essence and some people use the way of detoxifying and absorbing clots, but the composing prescription of our orally decoctions has the following characters: Firstly, the invigoration of more zang fu and multiple layers: we consider organism damages incurred by chemotherapy are many-sided, when it exist myelosuppression, the functions of spleen and stomach mostly impair and liver function may also impair. So, TiaoYinYing(TYY) therapy on myelosuppression in invigoration of liver, kindney, spleen and stomach, qi and blood etc, because liver stores blood, kindney stores vital essence, blood is changed by essence, spleen and stomach is source of qi and blood. Secondly, invigorating is combined with transporting and moving; TYY pays attention to prevent from abusing of invigoration and excessive invigoration, when using the way of invigorating, we also promote digestion and the running of qi and blood. Thirdly, the composing prescription of TYY also consider that TYY whether have therapeutic effects of tumor or not: strengthening body resistance to inhibit cancer, adjusting in one cast to anti cancer, this is also one aspect that chinese herbal compound is superior to simple western medicine. When we therapy on the myelosuppression patients which is caused by chemotherapy by using TYY in clinic, we always get significant curative effect. This chinese herbal compound is studied out by my tutor yaxian Liu who has evidence for traditional chinese medicine theory, analysises non- homogeneous organism damage which is caused by chemotherapy and combined with long-time clinical practice. This decoction is composed of ten herbs such as PrePared Rhizome of Relullannia, Cornus Officinalis, Chinese Yam, Galli Stomachichum Corium, Astragalus, Angelica Sinensis, Rhizoma Pollgonati, Tuberfleece flower, Salvia, Milettia Reticulata.Methods: 168 kunming mice of clean grade were randomly divided into 7 groups, i.e.,normal control group, model group, TYY group, Decomposed recipes of TYY , liuweidihuang decotion , decotion of eight ingredients, angelica decotion for replenishing blood. To replicate mice model of myelosuppression: each mouse of every group intraperitoneal injection CTX except for normal group, it were intraperitoneally given the same dose of saline continuous 3days. Intragastric administration after injecting CTX 4 hours was adopted once a day for 10 days.The normal and model groups were given the same dose of saline. The venous blood samples were taken for the measurement of peripheral hemogram on day 4, 7, 10 with full-automatic blood-cell counter. The 7th's blood used as detection of EPO at the same time.After that to fetch right femur to observe histomorphology(HE staining) and expression of GM-CSF(immunohistochemical method).To strip thymus and spleen in order to calculate thymus index and spleen index.The femur were stripped after drawing blood on day 10. To dash out bone marrow use PBS liquid and then to count the quantity of bone marrow nucleated cells.Results: 1.The effect onWBC:The WBC of model group decreased significantly compared with normal group in each period time. It droped to a lowest level on Day 4,then,was in an ascending tendency on Day 7 and 10.They were all lower than normal group and had significant differences compared with it (P<0.01). The WBC of every groups after treatment with TYY and other drug groups increased at different degrees in each period time. TYY ,Decomposed recipes of TYY and decotion of eight ingredients had significant differences compared with model group in the 4th day(P<0.05).The WBC of decomposed recipes of TYY,liuweidihuang decotion and angelica decotion for replenishing blood were higher than model group,but had statistical significance (P>0.05).However ,TYY and decotion of eight ingredients had significant differences compared with model group(P<0.05).Every drug groups could accelerate the rescovery of RBC in the 10th day and they had significant differences compared with model group (P<0.05).TYY was superior obviously to other groups(P<0.05). Every drug groups had significant differences compared with model group in the 10th day (P<0.05).TYY was superior obviously to other groups except for decotion of eight ingredients (P<0.01).2. The effect on RBC:The effect of mouse's RBC by CTX had certain regularity.The RBC showed downtrend after injecting CTX.It droped to a lowest level on Day 7, then, gradually recovered from the prophase depression. Decomposed recipes of TYY and liuweidihuang decotion had significant differences compared with model group on Day 4(P<0.05).Every drug groups resisted the decrease of RBC at different degrees in the 7th day.Among of them only TYY and Decomposed recipes of TYY had significant differences compared with model group(P<0.05). Every drug groups could accelerate the rescovery of RBC in the 10th day and they had significant differences compared with model group (P<0.05).TYY was superior obviously to other groups(P<0.05).3. The effect on HGB:CTX significantly decreased the HGB of mouse.It's tendency was the same as HGB. It droped to a lowest level on Day 7,then,gradually recovered from the prophase depression. Decomposed recipes of TYY and liuweidihuang decotion had significant differences compared with model group on Day 4(P<0.05). Every drug groups resisted the decrease of HGB at different degrees in the 7th day.Among of them only TYY, Decomposed recipes of TYY and liuweidihuang decotion had significant differences compared with model group(P<0.05). The HGB of every drug groups increased significantly in the 10th day and they had significant differences compared with model group (P<0.05).TYY was superior obviously to other groups(P<0.05).4. The effect on PLT:The PLT of Every groups by intraperitoneal injection of CTX were much lower than normal group in the 4th day (P<0.05 or P<0.01).It showed that the PLT of mouse were decreased succesfully by intraperitoneal injection of CTX.Only decotion of eight ingredients and TYY groups had significant differences compared with model group (P<0.05).Every drug groups resisted the decrease of PLT at different degrees in the 7th day.They had significant differences compared with model group except for liuweidihuang decotion (P<0.05). The PLT of Every groups increased further more in the 10th day.Only decotion of eight ingredients and TYY groups had significant differences compared with model group (P<0.05).The PLT of TYY were higher than decotion of eight ingredients,but had no differences between them.5. The effect on thymus index and spleen index: The thymus index and spleen index of model group were decreased significantly compared with normal group(P<0.05or P<0.01).Spleen index of every drug groups improved obviously after treatment with TYY and other drug groups. They were all have significant difference compared with model group(P<0.05 or P<0.01).TYY had no statistical significance compared with other drud groups(P>0.05).Only TYY group had significant difference compared with model group at improving thymus index.6. The effect on EPO:The EPO of model group was significantly higher than normal group.There were significant differences between these two groups (P<0.01). The EPO of blood were improved further more after treatment with TYY and other drug groups.TYY was superior obviously to other groups(P<0.05).7. The effect on nucleated cells in bone marrow:Nucleated cells in bone marrow of model group decreased significantly compared with normal group(P<0.05).Every drug groups could improve nucleated cells in bone marrow significantly and had significant differences compared with model group except angelica decotion for replenishing blood (P<0.05).TYY was superior obviously to other drug groups(P<0.05).8. Bone marrow hematopoietic cells of every group mouse were observed by HE staining. The normal group showed that:the structure of bone trabecula was clear, hematopoietic cells were abundant and arranged densely,bone marrow nucleated cells and megakaryocytes were more. The model group showed that: bone marrow tissues markedly necrosed and dropsied, sinus were dilated, bone marrow nucleated cells decreased significantly, megakaryocytes hadn't been seen. The Red Blood Cell spillovered and spindle fibrocyte proliferated. The bone marrow hematopoietic tissue of every groups rescovered in different degree after continuous using drugs 7 day. TYY group was more obvious. Sinus dilation wasn't obvious, bone marrow nucleated cells and megakaryocytes increased significantly, little spindle fibrocyte had been seen also.9. The effect on GM-CSF:The GM-CSF namely average optical density of model group were lower than normal group(P<0.05). The average optical density of every drug groups were higher than model group after treatment(P<0.05). TYY was superior obviously to other drug groups and normal group(P<0.05).Conclusions:1 TYY and every drug groups were all improve peripheral hemogram,bone marrow nucleated cells at different degrees. TYY was superior obviously to other groups at improving WBC aspect. It's superiority showed that taking effect quickly, improving amplitude largerly, keeping time longer;decotion of eight ingredients is next. Equivalent effects were found liuweidihuang decotion, angelica decotion for replenishing blood, decomposed recipes of TYY. TYY was best, liuweidihuang decotion and decomposed recipes of TYY were next at improving RBC and HGB aspects. TYY and decotion of eight ingredients were better in proving PLT aspects. They were superior obviously to model group. TYY was superior obviously to other groups at improving bone marrow nucleated cells.2 TYY and other drugs groups were all improving spleen index of myelosuppressed mouse. Only TYY group had significant difference compared with model group at improving thymus index. The reults suggested that TYY had Immunopotentiation and could improve extramedullary hematopoiesis.3 TYY and other drug groups were all promoting EPO secretion and expression of GM-CSF. TYY was superior obviously to other treatment groups .4 TYY and other drug groups were all promoting at different degrees resgovery of hematopoietic function of injured bone marrow. Sinus dilation wasn't obvious, bone marrow nucleated cells and megakaryocytes increased significantly. TYY was better than other groups and it was close to normal group.
