| Objective: based on the close relationship between the blood production of traditional Chinese medicine and the operation of yingwei,to explore the relationship between the operation of yingwei and the activation and regulation of hematopoietic stem cells.Methods: this study was divided into two parts: animal experiment and clinical experiment clinical experiment.The former detects the changes of hematopoietic regulation related factor OX?PGE2?NE?MT in vivo under different operation mode of Yingwei and drug intervention of Yingwei operation,the latter selects typical hematopoietic stem cell failure disease clinically,and observes the different clinical manifestations of Yingwei operation and Yingwei operation after drug intervention.Study 1: The healthy mice were randomly divided into two groups.The experimental group was given estazolam suspension to interfere with the operation mode of ying and wei,while the control group was given normal ying and wei operation mode.Peripheral blood and bone marrow fluid were collected to detect OX and PGE2 contents and analyze the differences.Study 2 :(1)The healthy mice were randomly divided into 4 groups,the zi and wu fang groups were given zi and wu prescription,respectively,and the zi and wu fang control groups were given normal saline.After single administration,peripheral blood and bone marrow solution were collected to detect NE and MT contents and analyze their differences.(2)The healthy mice were randomly divided into 6 groups.The time of positive and positive meridional prescription and negative meridional prescription and negative meridional prescription and negative meridional prescription and negative meridional prescription were respectively reversed,and the time of reverse meridional prescription and negative meridional prescription and negative meridional prescription were respectively reversed.In the control group,1 and 2 were respectively given normal saline at the corresponding time.(3)The healthy mice were randomly divided into 2 groups: meridional prescription combined group received meridional prescription corresponding to meridional prescription,and the combined control group received normal saline corresponding to meridional prescription.The contents of NE and MT were detected and analyzed for 6 consecutive days.Study 3: a day and a night were divided into four periods to observe the symptom scores offour clinical symptoms of dryness,fatigue,irritability and body heat in patients with chronic aplastic anemia at different periods and analyze the correlation between symptoms and time.Study 4: collect the first affiliated hospital of tianjin medical university onset of chronic aplastic anemia patients outpatient and ward,observe the meridian syndromes with the meridian syndromes before and after treatment,the peripheral blood leukocyte count,red blood cell count,hemoglobin concentration,platelet count,and make comparison before and after oneself,compare the clinical curative effect.Results:Study 1:OX and PGE2 in peripheral blood were significantly higher in the experimental group than in the control group,with significant statistical difference(P<0.05),while no difference was found in bone marrow fluid.Study 2:(1)The NE level in the peripheral blood prescription group was significantly lower than that in the control group,with significant statistical difference(P<0.05),and the MT level in the bone marrow solution prescription group was significantly higher than that in the control group,with significant statistical difference(P<0.05).(2)Compared with the control group,the NE level of the forward and reverse meridian prescription group and the sub-prescription group was higher,showing significant statistical difference(P<0.05).The MT level of the forward meridian prescription group was significantly higher,and the MT level of the forward sub-prescription group was lower,showing significant statistical difference(P<0.05).Comparison within the group: NE level in the forward and reverse groups and subgroups was significantly higher on day 3,6 and 9 than on day 0,with significant statistical difference(P<0.05).The NE level reached the peak on the3 rd day,showing a significant statistical difference(P<0.05).(1)the MT level of the forward and reverse direction group and the sub-direction group was significantly higher on day 3,6and 9 than that on day 0,showing a significant statistical difference(P<0.05).On the 6th day,MT level reached the peak with significant statistical difference(P<0.05).Compared with the day 0,MT level in the forward sub-square group increased significantly on day 6,showing a significant statistical difference(P<0.05).In the reverse direction group,compared with the day 0,the MT level on day 9 was significantly lower with significant statistical difference(P<0.05),and the MT level on day 12 was significantly higher with significant statistical difference(P<0.05).There was no significant difference in MT level in the reverse sub-squaregroup,and no statistical difference(P>0.05).(3)The MT level in peripheral blood of the meridional prescription combined group was significantly higher than that of the combined control group,with significant statistical difference(P<0.05).NE and MT levels in bone marrow solution were significantly higher than those in the combined control group,with significant statistical differences(P<0.05).Study 3: Clinical symptoms and time score of patients with chronic aplastic anemia:(1)in the dry symptoms,the score in the morning was higher than that in the other three groups,and there was a significant statistical difference compared with that in the evening and at night(P<0.05).(2)in terms of fatigue symptoms,the score in the afternoon was higher than that in the other three groups,and there was a significant statistical difference compared with that in the evening and at night(P<0.05).(3)among the symptoms of irritability,the score of night time period was higher than that of other three groups,and there was a significant statistical difference compared with other time periods(P<0.05).(4)among the symptoms of body heat,the score at night time was higher than that of the other three groups,with significant statistical difference(P<0.05).Study 4: After 3 months of treatment,after treatment of 18 cases in the ziwu decoction group,1 case was basically cured(5.6%),3 cases were relieved(37.5%),13 cases were significantly improved(72.2%),and the total effective rate was 94.4%.After treatment,1(6.25%)of the 16 patients in the non-meridian prescription group were basically cured,3(18.75%)were relieved,and 6(37.5%)were significantly improved.The total effective rate was 62.5%.The chi-square test showed that the total effective rate of the two groups was P < 0.05,showing a statistical difference.Compared with before treatment,red blood cells and hemoglobin were significantly increased in the meridional formulas group after treatment,with statistically significant difference(P<0.05),and red blood cells were significantly increased in the non-meridional formulas group after treatment,with statistically significant difference(P<0.05).Conclusion:Abnormal operation of ying and wei is manifested in different symptom changes of patients with slow recovery.The meridional prescription regulating the operation of ying and wei can improve its hematopoietic function,which may be exerted by influencing the activation factors of hematopoietic stem cells,OX,PGE2,NE and MT. |
