OBJECTIVE: To investigate and compare the expression of FHIT protein and TPX2 protein in lung cancer tissue and normal lung tissue, analyze the relation between FHIT protein,TPX2 protein and clinicpathological features(patient age,sex,smoking,genetral type,histological subtype,clinical TNM stage and lymphnode node metasasis),approach the role of FHIT protein and TPX2 protein in the pathogenesis and development of lung cancer in order to guide clinical diagnosis and evalute prognosis,and offer some theoretical evidences for the early diagnosis,prevention and therapy of lung cancer.METHODS: Real time quantitative(RT)-PCR and immunohisochemical SP were used to detect the expression of FHIT protein and TPX2 protein in lung cancer tissue and normal lung tissue .The results were analyzed by statistical software SPSS13.0.RESULTS: Real time quantitative(RT)-PCR revealed the expression level of FHIT gene was significantly lower in lung cancer tissue than in normal control lung tissue, the deletion rate of FHIT gene expression was 74.2%. The expression level of TPX2 gene was significantly higher in lung cancer tissue than in normal control lung tissue, the over-expression rate of TPX2 gene was 64.5%. Aberrant expression of FHIT gene was associated with smoking(P<0.05)and aberrant expression of TPX2 genehad no correlation with clinicopathological factors ( P>0.05 ) . Immunohistochemistry also showed the expression level of FHIT protein was significantly lower in lung cancer tissue than in normal control lung tissue, the deletion rate of FHIT protein expression was 67.7%. The expression level of TPX2 protein was significantly higher in lung cancer tissue than in normal control lung tissue, the over-expression rate of TPX2 protein was 70.9%. Aberrant expression of FHIT protein also was only associated with smoking(P<0.05). Aberrant expression of TPX2 protein was only associated with tissue differentiate level(P<0.05). There was no significant difference between FHIT protein and TPX2 protein(P>0.05).CONCLUSION:(1)There are low expression of FHIT protein and high expression of TPX2 protein in lung cancer. The abnormal expression of FHIT protein and TPX2 protein were associated with the occurrence and progression of lung cancer. They may be the important markers for tumor diagnosis. (2)The low-expression of FHIT gene was associated with smoking, indicated that abnormal FHIT gene expression may be the early molecular event, it may play a role in oncogenesis of lung and transformation to cancer, however, is not further deteriorative following disease advancing. (3)Excessive smoking may result in the changes of structure of FHIT gene, and induce lung cancer. (4)The over-expression of TPX2 protein was associated with tissue differentiate level. High-moderate in the differentiation of lung cancer over-expression of the positive rate was significantly higher than that of poorly differentiated lung cancer over-expression of the positive rate,it showed there was a significantly increasing trend along with the precancerous lesion of the over-expression of TPX2 protein. The over-expression of TPX2 protein probablely plays an very important roles in the tumorigenesis and progression of lung cancer. (5)No significant difference was found between the low-expression of FHIT protein and the over-expression of TPX2 protein.It demonstrates FHIT protein may not be in coordination with TPX2 protein in carcinogenesis and progression of lung cancer,both of them are independent factors.
