Objective: To investigate the correlation of the expression and clinicopathologic significance of FHIT,mdm2 in NSCLC.Methods: the expression of FHIT protein and mdm2 protein were detected by SP immunohistochemistry method,in which there were 53 cases of NSCLC and 19 cases of normal lung tissue,and the clinicopathological features of lung cancer were analyzed statistically.Results:1.The expression of FHIT was localized in the cytoplasm; the mdm2 was localized in the cytoplasm or karyoplast.2.Loss of FHIT expression in 53 cases of NSCLC was 62.26%,while 0 in 19cases of normal tissues(p<0.05). A marked reduction of FHIT protein expression was observed in patients with smoking history(70.73%)than that in patients without smoking history(33.33%)(p<0.05). Loss of FHIT expression in squamous cell carcinoma ( 81.82% ) ,with lymph node metastasis(74.19%) andⅢ~Ⅳstages(81.95%) was respectively higher than that in adenocacinoma(48.39%) ,without lymph node metastasis(45.45%) andⅠ~Ⅱstages(50%) significantly(p<0.05).The protein expression level of FHIT was reduced especially in poor (80%)or moderate differentiation(69.7%) ,which was much higher than well differentiation(20%)(p<0.05). Negative rate of FHIT protein expression was not significantly related to age and sex(p>0.05).3.Overexpression of mdm2 in 53 cases of NSCLC was 52.83%, while 0 in 19 cases of normal tissues(p<0.05). The excessive expression of mdm2 inⅢ~Ⅳstages(71.43%), with lymph node metastasis(67.74%)and poor differentiation(90%) was respectively higher than that inⅠ~Ⅱstages (40.63%), without lymph node metastasis (31.82%)and well(40%) or moderate differentiation(45.45%)(p<0.05). Positive rate of mdm2 protein expression was not significantly related to age, sex , smoking history and pathological types(p>0.05).4.There was negative correlation between expression level of the FHIT with mdm2 protein in NSCLC(p<0.05).Conclusion:1.There was significant difference between lung cancer tissues and normal tissues,in which the expression of FHIT protein was 100 percent. These results suggest that FHIT protein expression may occur at early stage of lung carcinogenesis and be associated with the oncogenesis and progression of cancer. Loss of the expression of FHIT protein was taken as one of the screening methods in earlier period of lung cancer among people with high risk factor.Negative rate of FHIT protein expression was significantly related to tumor pathological types, lymph node metastasis, pTNM stages, degree of cell differentiation and the smoking history of patients in NSCLC.2.Positive rate of mdm2 protein expression was significantly related to lymph node metastasis , pTNM stages and degree of tumor cell differentiation . The overexpression of mdm2 protein in lung cancer tissue may play an important role in the oncogenesis and development of lung cancer.3.Negative rate of FHIT expression and positive rate of mdm2 expression may be promote the malignant hyperplasy and metastasis of the tumor. Co-detected method of the expression levels of the two proteins in NSCLC can be important significance in analyzing the degree of malignancy,the potential capacity of metastasis and prognosis. Results they give remedy to help clinical doctor to cure the patients.
