Detection Of TGF-β Signaling Pathway-related Genes In SCLC | | Posted on:2010-05-22 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Xie | Full Text:PDF | | GTID:2144360275975754 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Background.Transforming growth factorβ(TGF-β) is a multifunctional cytokine that plays a central role in the signaling networks that regulate cell growth,differentiation, adhesion,apoptosis,migration and modification of the microenvironment in which the cells reside.TGF-βexerts its function in intracellular signaling through its receptors complex.The ligand,receptor,Smad protein and target gene regulating the transription compose the depressing pathway.Every factor's dysfunction in this pathway will lead to the derangement of this pathway,lossing control of the cell growth differentiation,then promote the cancer's occurrence and development.In the past study,they found that TGF-βLPHN-2 and DACH1 may be the factors in this pathway.For the research about these genes is almost blank especially in SCLC in the world,we now inspect their expression in the SCLC.Objective:To study the TβRⅡ,LPHN-2 and DACH1 expression in SCLC.Combined with its expression in NSCLC we explore the relationship between their expression and pathological types.Research Materials and Methods:In this study In the present study,27 formalin-fixed, paraff'm-embedded SCLC cases and their corresonding normal tissue were collected by the Laboratory of the Department of Thoracic Surgery,Changhai Hospital.65 formalin-fixed, paraffin-embedded NSCLC cases were collected to be the control group.Then we use immunohistochemical staining method to detect the expression.And score according to their staining intensity and staining proportion.Statistic analysis was performed with chi-square test(x~2 test),corrected x~2 test or Fisher exact test.A P-value of less than 0.05 was considered statistically significant.Result:Inthe present study,62.96%(17/27) of tum0r tissues showed reduced(the score wasl—6) or loss(elg.negative) of TβRⅡexpression in SCLC and 35.38%(23/65) of tumors showed reduced(the score wasl—6) or loss(e.g.negative) of TβRⅡexpression in NSCLC.Among the NSCLC conclude 9 Sq-c and 12 Aq-c.59.26%(16/27) of tumor tissues showed reduced(the score was 1—6) or loss(e.g.negative) of LPHN2 expression in SCLC and 24.62%(16/65) of tumors showed reduced(the score wasl—6) or loss(e.g. negative) of TGFBR2 expression in NSCLC.Among the NSCLC conclude 8 Sq-c and 7 Aq-c.74.07%(20/27) of tumor tissues showed reduced(the score wasl—6) or loss(e.g. negative) of DACH1 expression in SCLC and 43.08%(28/65) of tumors showed reduced (the score wasl—6) or loss(e.g.negative) of DACH1 expression in NSCLC.Among the NSCLC conclude 9 Sq-c and 15 Aq-c.There is no association existed between the TβRⅡ,LPHN-2 and DACH1 expression and the clinical features.In TβRⅡ,the frequency of reduced type in SCLC is higher than that of adenocarcinoma and squamous cell carcinoma,and NSCLC(0.01 | | Keywords/Search Tags: | SCLC, NSCLC, TGF-β, TβRⅡ, LPHN-2, DACH1, immunohistochemistry | PDF Full Text Request | Related items |
