Objective: Now there are many scientific researching reports about how to use acupuncture therapy to cure the disease of cerebral ischemia. Besides, the reserachers have accumulated so much experience, it has great value in clinical application, but only a few reports on relationship between acute cerebral ischemia reperfusion damage BBB and acupuncture therapy protect BBB. This topic is researching about whether acupuncture therapy is good for BBB after acute cerebral ischemia reperfusion by needling Baihui and Shuigou acupoints and moxibusting Zusanli on rats. To observe the BBB permeability change on some biochemical-indicators (S100β,NO,SOD)which have grerat effect to BBB' s permeability. From this experiment,we will do further study about the principle why acupuncture and moxibustion therapy can cure the damaged BBB after cerebral ischemia reperfusion.Methods: Totally 96 healthy SD rats were selected and divided randomly into 4 groups, each group have 24 rats. The groups included normal group,sham-operated group,model group and electro-acupuncture and moxibustion group. Each guoup has 4 phase, it contains 6 hour phase , 12 hour phase, 24 hour phase, 48 hour phase. Each phase have 6rats. The model of ischemia reperfusion of cerebral middle artery was established by the modified thread occlusion method. The electro-acupuncture and moxibustion group needling Baihui and Shuigou acupoint and moxibusting Zusanli after the model is successful, and then give another 30 minutes curing before to draw the materials from rats ( acupuncture 2 time in all). Drawing blood from the eyeball of the rat and use heparin to prevent clotting, then use hydroextractor 4000 rol1/min centrifugating 10 minutes, conservation the blood-serum into cold-storage within -20℃in order to survey and evaluate 3 biochemical-indicators (NO, S100β, SOD). Choose 2 rats in each phase decollated directly to extract unsalted brain tissue by TTC colouring then computing the infarct size and carring out statistical treatment. The last 4 rats in eatch phase also decollat directly to extract unsalted brain tissue and obtain the part which blood-suppling on brain in to 2. 5% glutaral, then use H-600 transmission electron microscope carring out the image analysis. The entire datum was analyzed by statistics.Results:1.MCA0 and reperfusion injury can damage the rats activity distinctly. The nerve symptoms of electro-acupuncture and moxibustion group was ameliorated obviously; there was a significant difference between model group and electro-acupuncture and moxibustion group. (P<0. 05).2. There was visible infarct focus in some ischemia area after focal cerebral ischemia-reperfusion by TTC colouring. Compared with model group, the methods of electro-acupuncture and moxibustion can moderated and lighten cerebral infarct volume (P<0. 05). There also have visible effect between 6h phase in electro-acupuncture and moxibustion group and 24h phase in electro-acupuncture and moxibustion group, the conclusion has statistical significance.3. ELISA and NO experiments showing S100βand NO in model group are raising up in blood serum after cerebral ischemia-reperfusion compared with electro-acupuncture and moxibustion group (p<0. 05). There is no significant difference between normal group and electro-acupuncture and moxibustion group (p>0.05). Not statistically significant. The SOD contents in model group are lower than electro-acupuncture and moxibustion group (p<0. 05) Conclusion: The NO,S100β, contents in model group are lower than electro-acupuncture and moxibustion group (p<0. 05). This means cerebral ischemia-reperfusion can damaged BBB, increasing the BBB' s permeability and making some micromolecule substance which may harmful in blood passing the BBB system. The permeability of the BBB can be degraded by using electric acupuncture and moxibustion. The curatio can be also change the microcirculation and protect the BBB system. by using electric acupuncture and moxibustion. The SOD contents in model group are lower than electro-acupuncture and moxibustion group (P<0. 05) hint brain tissue oxygen deficiency is aggravated after cerebral ischemia-reperfusion but can be degraded by using electric acupuncture and moxibustion. From the 4 phase in electro-acupuncture and moxibustion group, we can find the therapeutic efficacy in 6 hour and 12 hour is better than 24 hour than 48 hour. (P<0. 05) This indicates that the clinical ability to save those who have not timely changed cerebral ischemia and reperfusion necrosis of the surrounding "penumbra" brain tissue, is currently the treatment of cerebral ischemia-reperfusion injury of the principal means.
