Background and Objective Ischemic cerebrovascular disease is a common disease which occurrs frequently.How to make the rescue and treatment of acute ischemic stroke more effectively,minimizing the rate of disability,improving patients's quality of living has become the world's important medical issues which has been attracted more concern.traditional view holds the opinion that make the ischemic areas referfusion as soon as possible in oder to rescue the dying neurons,glial cells and neural endothelial cells are the key course of treatment of ischemic cerebrovascular disease.however,long-term' s hypoxia cause the brain' s microenvironment to change;a large number of anaerobic glycolysis caused by the accumulation of lactic acid;prolonged ischemia cause mitochondrial destroyed;energy depletion of the body and so on.these changes bring about interaction between the restored blood flow and damaged organizations,resulting ischemia reperfusion injury.In order to resolve this contradiction,through a large number of researches,researchers found that ischemic area can start their own compensatory angiogenesis after cerebral ischemia,in order to compensate only a part of blood supply,but not the whole. In recent years,with an in-depth study in the molecular level of ischemic cerebrovascular disease,people found a variety of cytokines involved in angiogenesis proces and angiogenesis proces 's control and adjustment.So far,vascular endothelial growth factor(VEGF)is seen to play a key and special role in angiogenesis proces,and hypoxia-inducible factor 1α(HIF-1α) have.great significanses in mediate signal comitte betwwen hypoxia and angiogenesis,which can be activated by hypoxia and help to induce the expression of VEGF.Adjust the expression of angiogenesis-related cytokines,promote angiogenesis proces and restore the blood supply of ischemic areas can help restore the function of neurons after cerebral ischemia.A large number of clinical reports have confirmed that acupuncture is a effective treatment of ischemic stroke.My subject,used the middle cerebral artery occlusion models,selected "BAIHUI","SHUIGOU","HOUSANLI", through electro-acupuncture treatment,to observe some changes in the expression of VEGF and HIF-1αin MCAO rats hippocampus after focal cerebral ischemia and explore electro-acupuncture treatment 's role in promoting angiogenesis and its possible mechanism, in order to provide a experimental basis.Methods SD rats were randomly divided into the normal control group,sham operation group,model group(isehemia 30 min reperfusion 12h,1day,2days),electro-acupuncture treatment group(ischemia 30 min reperfusion 12h,1day,2days).The former two groups have 10 in each group,the latter two groups have 10 in each sub-group.Used the suture in MCA to establish MCAO isehemia-reprfusing model.The remarks of nerve function deficit referring to Lundy remarking standards.The acupoints of "BAIHUI","SHUIGOU"and"HOUSANLI" were selected.Electro-acupuncture treatment group take the first treatment 1h after isehemia-reprfused,and every 12-hour electro-acupuncture for 30 minutes,the last treatment at 1 hour before kill it.When ended the treat,according to the phase,take rat' Hippocampus,Make paraffin-embedded,sections,each wax block cut 3 pieces of 5μm section in hippocampus.Use immunohistochemical S-P method,observe microvascular density and positive expression of HIF-1αand VEGF by optic microscope.the groups were observed in hippocampus of rats with ischemic microvascular density and,all the data are input into the computer, analyzed with SPSS11.0 software statistical package.The experimental results of microvessel density are expressed by average士standard deviation,while the positive rate of VEGF,HIF-1αcompared with x~2.Results(1) Microvessel Density:Microvessel density in hippocampal of every model sub-group group are higher than the normal group and sham group,Microvessel density in hippocampal of every electro-acupuncture sub-group are higher than the corresponding phase of model group,there is significant difference, (P <0.01).(2) Expression Of VEGF,HIF-1α:Rats in normal group and sham group only have a very small number of positive cells in hippocampus,while the positive cells increased significantly in model group,and electro-acupuncture group expression in hippocampus(p <0.05).Positive expression rate of VEGF in model group and electro-acupuncture group are 78.6%,84.6%respectively; while HIF-1αare 71.4%,92.3%respectively,the difference has statistically significant(p <0.05);(3) The Comparison Of Expression Of VE6F And HIF-1αIn every electro-acupuncture sub-group group:Positive expression- rate,of VEGF in group of electro-acupuncture 12h,1d,2d are 60%,87.5%,87.5% respectively,while HIF-1αare 80%,87.5%,87.5%respectively.This shows that expression of VEGF,HIF-1αale increasing gradually with the treatment time increased,but the differences between the groups does not have statistically significant(p> 0.05),this may be associated with a small number of cases.Conclusions Electro-acupuncture treatment based on the acupoints of "BAIHUI","SHUIGOU"and"HOUSANLI" is effective for treat cerebral ischemia-reperfusion injury,its mechanism may be relative to the promotion of expressions of HIF-1αand VEGF in hippocampal area and regulate the angiogenesis process after cerebral ischemia-reperfusion.
