Objective:Ischemic cerebrovascular disease (ICVD) , also known as ischemic apoplexy, belongs to the category of the traditional chinese medicine's wind stroke, has the characteristic of high incidence of a disease,high case fatality,high incidence of mutilation. ICVD is one of the most clinic familiar cerebrovascular disease, badly intimidates people's health and lives.Brain tissue is very sensitive to ischemia, after ischemic, it will soon necrose in local cerebral ischemia center region, but in penumbral field nerve' function incapacitate or nearly incapacitate and nerve cells still survival.Now people have realized that the key to improve ICVD's curative effect is to ameliorate blood accommodation in ischemic region, saving nerve cell at ischemic penumbra. So treating ICVD, the most important means and aim is that promoting blood vessel renascence and forming branch blood vessel to resume blood accommodation in time and nourish nerve cell at ischemic penumbra. Nowaday, scientific study indicate the process of angiogenesis is strictly controlled by some advance factors and some control factors, the balance or unbalance of which decide the growth state of blood vessel. And, vascular endothelial growth factor (VEGF) is the best advance factors, endostatin (ES) is the one of the best control factors. So, the content of VEGF and ES can react angiogenesis's condition. To ICVD, acupuncture is the effective method in clinic. Some researches have approved that acupuncture can accelerate angiogenesis, increase tiny blood vessel's density, but the mechanism need deeply study.This research adopt the model of middle cerebral artery occlusion (MCAO) , electroacupuncture (EA) in baihui,shuigou and zusanli points, to observe the effects of EA on VEGF and ES expression around the infarction locus in rats with focal cerebral ischemic reperfusion (CIR) , discuss possible mechanism of EA promoting angiogenesis, and more study the mechanism of acupuncture treating ICVD, provide clinic with laboratorial and academic evidence.Method:According to the time of CIR, one hundred SD rats were randomly divided into 10 groups: normal group,pseudo-operation group,model A group,model B group,model A group,model A group,EA A group,EA B group,EA C group,EA D group .The focal CIR models were established by improved Zea-Longa method and the expression of VEGF and ES were observed with immuneohistochemical method.Result:1. VEGF and ES both express in low level at cerebrovascular endothelium cell in normal group and pseudo-operation group. Comparing two groups, P>0.05.2. Compares with normal group and pseudo-operation group, VEGF and ES both highly express in model group, P<0.01.3. Longer the time of CIR, lower the expression of VEGF and ES in model group and EA group.4. Compares between model group and EA group, at the same time, VEGF increases expression and P<0.01 except C group in which P>0.05; and ES lower expression, P<0.01.Conclusion:1. An ecumenical operation hurt has little influence in VEGF and ES.2. Lacking blood and oxygen are the important factor in spur VEGF's expression. The promoted expression of ES is likely associated with the protected mechanism which can prevent the excess of angiogenesis.3. Ischemia reperfusion-1d is the most likely time that the expression of VEGF and ES come to the peak value.4. EA can increases VEGF's expression and decline ES's expression, which will be one of the possible mechanism for EA accelerate angiogenesis.
