Objective:Cerebrovascular disease(Cerebral Vascular Disease,CVD) is harmful to health of human life and is the common and frequently-occurring disease,so its morbidity,mortality and disability are high.Brain tissue are so particularly sensitive to ischemia that after ischemia,the center within minutes of regional ischemia neuronal damage may be irreversible;and around the zone of the brain tissue there is collateral circulation to support rapidly after appropriate treatment can be saved,referred to as " ischemic penumbra(Ischemic penumbra,IP) ",the zone of ischemic brain tissue after the loss of electrical activity,but to maintain membrane voltage and transmembrane potential is an early pathological basis of stroke treatment.To improve the penumbra of brain tissue angiogenesis and collateral blood vessel formation and in time to resume penumbra region the blood supply to save the protection of reversible penumbra neurons is the treatment of ischemic cerebrovascular disease and one of the purposes of an important means.Vascular endothelial growth factor (vascular endothelial growth factor,VEGF) is considered the most potent, highest-specific factors into a blood vessel;angiopoietin(angiopoietin,Ang) is the first identified with the role of angiopoietin cytokines.Acupuncture is an effective means of the clinical treatment of ischemic cerebrovascular disease, research has confirmed that acupuncture can promote angiogenesis in ischemic areas,an increase of microvessel density,but its mechanism remains to be further study.Theexperiment use the focal cerebral ischemia-reperfusion rat model, electro-acupuncture "Baihui","Shuigou","Housanli" Point to observe the electrical response to cerebral ischemia-reperfusion in rat focal side(right) large in the hippocampus of VEGF,Ang-1,CD34,will investigate the electro-acupuncture to promote angiogenesis in ischemic areas of the possible mechanism,further in-depth study role of mechanism of acupuncture treatment of ischemic cerebrovascular disease,so as to clinical acupuncture treatment of ischemic cerebrovascular disease to provide a basis for the experiment.Method:Healthy adult SD rats,80,male and female informal,were randomly divided into normal control group,sham-operated group,model group,EA group;Among them,the model group,respectively,electro-acupuncture group after ischemia-reperfusion at different times(12h,1d,2d),divided into A,B, C three subgroups.Modified suture prepared rat focal cerebral ischemia and reperfusion(MCAO) model,in the light of Zea-Longa score standard neurological score,for screening of animals.Electro-acupuncture "Baihui", "Shuigou","Housanli" point,rats with lesions detected by expression of VEGF,Ang-1,CD34 of the side(right) in hippocampal brain by the way of immunohistochemistry(SP).Result:1.The normal group compared with sham-operated group VEGF,Ang-1, CD34 in the brain vascular endothelial cells showed low expression or no expression;no significant difference between the two groups(P>0.05).2.The model group and normal group,compared with sham group, VEGF,Ang-1,CD34 expression was significantly enhanced,with significant difference(P<0.05).3.In 12h,1d,2d phase,expression of VEGF,Ang-1 the model group or electroacupuncture group with the number of CD34 expression in microvascular have some relevance.4.Electro-acupuncture group compared with the model group,VEGF expression,with the exception of electro-acupuncture group C compared with Model C group(P>0.05),no significant difference,the remaining phase of the corresponding comparison group are significant differences(P<0.05);Ang-1 expression was significantly increased,and the corresponding phase group,are significantly different(P<0.05);CD34 microvessel expression,the corresponding phase group,no significant differences.Conclusion:1.The normal group with sham-operated group VEGF,Ang-1,CD34 in the brain vascular endothelial cells showed low expression or no expression; no significant difference between the two groups(P>0.05).2.The model group and normal control group,compared with sham group,VEGF,Ang-1,CD34 expression was significantly enhanced,with significant difference(P<0.05).3.In 12h,1d,2d phase,the model group,electroacupuncture group and the group VEGF,Ang-1 expression and the number of CD34 expression in microvascular have some relevance.4.Electro-acupuncture group compared with the model group,VEGF expression,with the exception of electro-acupuncture group C compared with Model C group(P>0.05),no significant difference,the remaining phase of the corresponding comparison group are significant differences(P<0.05);Ang-1 expression was significantly increased,and the corresponding phase group,are significantly different(P<0.05);CD34 microvessel expression,the corresponding phase group,no significant differences.
