Zi Jinsha is the dry root of Angelica polymorpha Maxim.of umbelliferae plant.It is a folk medicine which is applied to treat stomachache in Tujia Nationality.Angelica polymorpha Maxim.has the efficacy of wind-expelling, dampness-eliminating,cold-dispelling,and pain-alleviating,which is of significant active effect on treating stomachache,bellyache,stethalgia, rheumatoid joint pain and so on.It is one of medicinal plants which are worthy of researching and developing.In this thesis,chemical constituents of Angelica polymorpha Maxim. were systematically investigated.Six compounds were isolated by silica gel chromatography,recrystallization and Sephadex LH-20 chromatography.On the basis of physical and chemical properties as well as detail spectroscopie analysis(IR,MS,1H-NMR,13C-NMR,DEPT),five compounds have been identified as:isoimperatorin,bisabolangelone,oxypeucedanin,oxypeucedanin hydrate,β-sitosterol.On the basis of chemical constituents investigated,preparation of the standard substance of Angelica polymorpha Maxim.was carried out.Two compounds were isolated by silica gel chromatography,recrystallization, Sephadex LH-20 chromatography and semi-Preparative HPLC.By employing physical and chemical properties,modern spectrometric analysis techniques such as(MS,1H-NMR,13C-NMR,the structures of two compounds had been elucidated as isoimperatorin and bisabolangelone.The purity detected by TLC and HPLC was in line with the requirements of standard substance for determination.By screening of methods extraction,solvent extraction and developing solvent,the TLC condition was founded to analysis the crude character and quality.The mobile phase was chloroform ethyl acetate(96:4). An UV spectrophotometry for determining total coumarins in Angelica polymorpha Maxim.was established.Isoimperatorin had been chosen as the reference substance for quantitative determination of total coumarins in five samples.The content of total coumarins in the crude was initially established as not less than 1.8%.The UV spectrophotometry was suitable for the determination of total coumarins.The RP-HPLC method was used to determine the content of isoimperatorin and bisabolangelone in Angelica polymorpha Maxim.The analysis was carried out on Kromasil 100A C18 column(250mm×4.6mm,5μm).The mobile phase was acetonitrile-water with gradient elution.The flow-rate was 1.0mL/min and wave-length at 254nm and temperature at 25℃.The minimum content of isoimperatorin should be 0.23%,and bisabolangelone should be 0.22%.The content of isoimperatorin and bisabolangelone in the crude can be determined at the same time.The content of volatile oil,moisture,total ash,acid-insoluble ash and extractive in Angelica polymorpha Maxim.was determined according to the 2005 edition of Chinese Phannacopoeia appendix method,and the standard was initially formulated.The method is accurate,reliable,reproducible.In this thesis,the chemical constituents and quality standard were systematically investigated.On the basis of above experimental results,quality specification draft and additional remarks not detailed in draft was worked out. It provides the scientific bases for the quality of Angelica polymorpha Maxim. in evaluation criterion.Prominent application perspective would emerge in the respect of utilization on the rich resources of Angelica polymorpha Maxim.in the future...
