Meicha is made by the tender stems and leaves of A.megalophylla Diels et.Gilg.It is used to treat hypertension in clinic and has a remarkable efficacy. The main effective chemical constituents are total flavonoids,which are proved by pharmacology experiments that can reduce high blood pressure,high blood lipid,glycemia and can protect liver,improve immunity.Furthermore,its adverse effect is low.So it can be produced as a new effective drug of treating Cardiov-ascular diseases.On the chemical constitution study of Meicha and preparing Meicha total flavonoid extracts study,we study quality control of Meicha total flavonoid extracts,which establish certain base for the new drug study of Meicha.First,we collect some correlative data and literature of Meicha to Determine Meicha's effective chemical ingredients,pharmacology effect and chemical quality,which direct the way of how to study on quality control of Meicha total flavonoid extracts.The quality and character of Meicha total flavonoid extracts are examined;Ampelopsin and Myricetin of Meicha total flavonoid extracts are identified by silica gel thin-layer chromatography and polyamide thin-layer chromatography;The contents of the total flavonoids,Ampelopsin and Myricetin of Meicha total flavonoid extracts are measured;study on HPLC fingerprint of Meicha total flavonoid extracts;the contents of residual organic solvents in Meicha total flavonoid extracts are measured.Result:Meicha total flavonoid extracts are yellow amorphous powder;Meicha total flavonoid extracts are slightly soluble in pH1 hydrochloric acid solutions,Ph6.8 phosphate buffer and distilled water,and are easily soluble in ethanol;the angle of repose of Meicha total flavonoid extracts is 35.7±0.40°;the CRH of Meicha total flavonoid extracts is about 82%;the density of Meicha total flavonoid extracts is 0.68 mg·mL-3;the contents of water of Meicha total flavonoid extracts conform to the requirement;identifying Ampelopsin and Myricetin with silica gel thin-layer chromatography and polyamide thin-layer chromatography are separately established; the contents of the total flavonoids,Ampelopsi and Myricetin are measured, and are not lower than 78%,44%,11%respectively.HPLC fingerprint of Meicha total flavonoid extracts is established by study on 10 batches of Meicha total flavonoid extracts,showing 10 common peaks and similarity>0.9,peak 3,9 and 11 as Ampelopsin,Myricetin and Quercetin;the residue of Divinylbenzene is less than 0.002%,and Benzene,Toluene,Xylene,Styrene and n-Hexane are not respectively detected in Meicha total flavonoid extracts.Research methods of quality control of Meicha total flavonoid extracts are set up through the study of the subject.
