ObjectiveThis issue through the establishment of the large intestine damp-heat in the rat model to study UC Qing-chang-tang of the large intestine damp-heat in the treatment of UC and its mechanism,in particular through the observation of the large intestine damp-heat in UC rats,in-depth treatment of the large intestine reveals heat soup Qingchang UC's role-based mechanism for the treatment of UC in Chinese medicine to provide a foundation of experimental and theoretical basis.MethodsWill be 40 healthy SD rats(male and female half) by male and female were randomly divided into 5 groups,namely A(normal control group),B group(model group),C group(SASP group),D group(Qingchang soup conventional dose group) and E group(Qingchang soup of high-dose group), 8 in each group.The use of dextran sulfate sodium diet add add acid environment with complex law,the large intestine damp-heat in rat model of UC.Model after the completion of drug administered to the treatment of rats: normal group and model group were administered distilled water;Qingchang conventional dose group and soup soup Qingchang given high-dose group were administered the treatment Qingchang Tang,;SASP group SASP treatment with oral suspension,were administered for more than 14d.Result 1 Qingchang Decoction on rat large intestine damp-heat in the colon of UC morphologyQingchang soup in each group and SASP group UC rat colonic mucosa with mild congestion and edema,no erosion,ulcers,endoscopic mucosal ulcer-free,within the lamina propria there is a small number of lymphocytes, plasma cells,eosinophil infiltration,colon general concept of mucosal injury and injury of scores of colonic mucosa compared with the model group has statistical significance.Qingchang soup can reduce colon tissue damage and promote repair of lesions UC colon,large intestine damp-heat in the treatment of UC and effective.2 Qingchang Decoction on rat large intestine damp-heat in UC colon mucosa damage index(CMDI points) the impact ofSASP group after treatment,conventional dose group Qingchang soup and soup Qingchang colonic mucosa of animals large doses significantly reduced congestion and edema.SASP,Qingchang soup soup Qingchang conventional dose and high dose rats three CMDI(points) than the normal control group was significantly higher(P <0.01).After treatment,SASP, Qingchang soup soup Qingchang conventional dose and high dose rats three CMDI(hours) was significantly lower with the normal control group showed no significant difference between groups(P> 0.05);compared with the model group were significantly different(P <0.01)3 Qingchang Decoction on rat large intestine damp-heat in UC intestinal tissue cytokine(TNF-α,IL-1 and IL-6) expression ofDescription soup with Chinese medicine Jianpi Yiqi Qingchang, enhance the defense capabilities of the body,reducing pro-inflammatory factor TNF-α,IL-1 and IL-6 expression and regulation of immune function, reducing the role of colonic mucosa damage.4 Qingchang Decoction on rat large intestine damp-heat in UC intestinal tissue NF-κB p65,TLR4 expressionQingchang soup each dose group after treatment compared with the model group:rat colon tissue NF-κB p65,TLR4 expression declined.Qingchang soup that has a lower NF-κB p65,TLR4 expression,thereby reducing cytokine secretion,reducing the role of colonic mucosa damage.ConclusionsNF-κB p65,TLR4 and cytokines TNF-α,IL-1 and IL-6 may be involved in the large intestine damp-heat in the occurrence of UC and development. Qingchang Decoction foru UC rat large intestine damp-heat in the mechanism is likely to be through inhibiting the expression of TLR4 protein,blocking its receptor protein,phosphorylation of NF-κB activation function,thereby reducing the secretion of pro-inflammatory factor to injury endothelial cells to repair,then the large intestine damp-heat treatment to achieve the purpose of UC.
